Crystal structure of the nicotinamidase/pyrazinamidase PncA from Bacillus subtilis
Biochemical and Biophysical Research Communications, 2018The nicotinamidase/pyrazinamidase PncA is a member of a large family of hydrolase enzymes that catalyze the deamination of nicotinamide to nicotinic acid. PncA also functions as a pyrazinamidase in a wide variety of eubacteria and is an essential coenzyme in many cellular redox reactions in living systems.

Molecular characterization of the Borrelia burgdorferi in vivo-essential protein PncA [PDF]
The conversion of nicotinamide to nicotinic acid by nicotinamidase enzymes is a critical step in maintaining NAD+ homeostasis and contributes to numerous important biological processes in diverse organisms. In Borrelia burgdorferi, the nicotinamidase enzyme, PncA, is required for spirochaete survival throughout the infectious cycle.

Characterization of pncA mutations in pyrazinamide-resistant Mycobacterium tuberculosis
Antimicrobial Agents and Chemotherapy, 1997Pyrazinamide (PZA) is a first-line drug for short-course tuberculosis therapy. Resistance to PZA is usually accompanied by loss of pyrazinamidase (PZase) activity in Mycobacterium tuberculosis. PZase converts PZA to bactericidal pyrazinoic acid, and the loss of PZase activity is associated with PZA resistance.

Drug resistance mechanism of PncA inMycobacterium tuberculosis
Journal of Biomolecular Structure and Dynamics, 2013Tuberculosis continues to be a global health threat. Pyrazinamide (PZA) is an important first-line drug in multidrug-resistant tuberculosis treatment. The emergence of strains resistant to PZA represents an important public health problem, as both first- and second-line treatment regimens include PZA. It becomes toxic to Mycobacterium tuberculosis when
