Polo-like kinase inhibitor BI2536 induces eryptosis
Wiener Medizinische Wochenschrift, 2022BI2536 is potent inhibitor of polo-like kinases PLK1, 2, and 3. The inhibition of PLKs in nucleated cells induces apoptosis by perturbing the cell cycle with consequent engagement of mitotic catastrophe. BI2536 is being tested as chemotherapy in various phase I/II/III clinical trials.

Polo-like kinase inhibitors in hematologic malignancies
Critical Reviews in Oncology/Hematology, 2016Polo-like kinases (Plk) are key regulators of the cell cycle and multiple aspects of mitosis. Two agents that inhibit the Plk signaling pathway have shown promising activity in patients with hematologic malignancies and are currently in phase III trials.

Discovery of thiophene inhibitors of polo-like kinase
Bioorganic & Medicinal Chemistry Letters, 2009The discovery and development of a series of thiophenes as potent and selective inhibitors of PLK is described. Identification and characterization of 2, a useful in vitro PLK inhibitor tool compound, is also presented.

Polo-like kinases and the orchestration of cell division
Nature Reviews Molecular Cell Biology, 2004Polo-like kinases (Plks) are increasingly recognized as key regulators of mitosis, meiosis and cytokinesis. In agreement with a broad range of proposed functions during cell division, Plks are subject to complex temporal and spatial control. Recent findings are uncovering the mechanisms of Plk regulation, notably their targeting to different cellular ...

Polo-like kinases in cell cycle checkpoint control
Frontiers in Bioscience, 2003Recent studies from various eukaryotic model systems indicate that polo-like kinases (Plks) play an ever-increasing role in the regulation of cell cycle progression. Early genetic studies have demonstrated that Cdc5, a budding yeast counterpart of vertebrate Plks, is essential for mitosis.
