Diversity of Pharmaceuticals Enhances Antibiotic Resistance in the Invertebrate Gut via Biofilm‐Mediated Mechanisms

open access: yesAdvanced Science, EarlyView.
Pharmaceutical diversity acts as an independent driver of antibiotic resistance in soil invertebrates. While bulk soil remains unaffected, the collembolan gut microbiome exhibits significant resistance gene enrichment under complex chemical exposure and diurnal warming.

Evaluation of the Clinical Systems for Polymyxin Susceptibility Testing of Clinical Gram-Negative Bacteria in China

open access: yesFrontiers in Microbiology, 2021
ObjectivesThe performance of mainstream commercial antimicrobial susceptibility testing systems on polymyxins has not been well evaluated in China. In this study, three antimicrobial susceptibility testing systems were evaluated for polymyxin B and ...

Multi‐Targeting Carnosic Acid Kills Drug‐Resistant Helicobacter pylori With Narrow‐Spectrum Activity

open access: yesAdvanced Science, EarlyView.
Carnosic acid selectively eradicates drug‐resistant Helicobacter pylori through a multi‐targeted mechanism involving urease inhibition, membrane disruption, and biofilm eradication. It exhibits no detectable resistance, potent in vivo efficacy, and minimal impact on gut microbiota, positioning it as a promising narrow‐spectrum lead compound against ...

Polymyxin Delivery Systems: Recent Advances and Challenges

open access: yes, 2020
Polymyxins are vital antibiotics for the treatment of multiresistant Gram-negative ESKAPE pathogen infections. However, their clinical value is limited by their high nephrotoxicity and neurotoxicity, as well as their poor permeability and absorption in ...

Identification of a Nonribosomal Peptide Analog With Activity Against Multiple Gram‐Positive Bacteria via a Synthetic Bioinformatic Natural Product Discovery Approach

open access: yesAdvanced Science, EarlyView.
The genome of Rhodococcus erythropolis D‐1 was analyzed by bioinformatic tools to mine a novel nonribosomal peptide synthetase (NRPS) gene cluster. A nonribosomal peptide analog ZURJC5 associated with the NRPS was chemically synthesized. Through structure‐activity relationship studies, ZURJC28 was ultimately obtained and showed antibacterial activity ...

Detection assay of polymyxin resistance coding mcr-1 gene based on CRISPR/Cas13a system

open access: yesFrontiers in Cellular and Infection Microbiology
IntroductionPolymyxins are reserved as an ultimate defense against multidrug-resistant bacteria. The emergence of the polymyxin resistance gene mcr-1 poses a potential risk for the treatment of severe infections caused by Gram-negative bacteria.

Targeting Lipopolysaccharide Transport Induces Membrane Lipid Remodeling and Sensitizes Acinetobacter baumannii to Colistin Treatment

open access: yesAdvanced Science, EarlyView.
This study identifies C4 as a lead inhibitor of the Lpt system. Notably, C4 potentiates colistin activity by disrupting LPS transport and remodeling phospholipid homeostasis, revealing a functional interplay between the Lpt and Mla systems. These findings establish a mechanistic link between Lpt inhibition and membrane lipid remodeling, positioning Lpt–

Polymyxin B binding sites in Escherichia coli as revealed by polymyxin B-gold labeling.

open access: yes, 1987
A complex of polymyxin B, bovine serum albumin, and colloidal gold was prepared and used for the ultrastructural localization of polymyxin B binding sites on thin sections of Epon-embedded Escherichia coli cells.
