AbstractGuanosine 5′‐(tri)diphosphate, 3′‐diphosphate [(p) ppGpp] is a small nucleic acid that helps bacteria survive in limited environments. Gene chip shows that (p) ppGpp is a global transcription‐regulator of genes related to important bacterial metabolic processes.

Studies on the in vitro synthesis of ppGpp and pppGpp
Biochimica et Biophysica Acta (BBA) - Nucleic Acids and Protein Synthesis, 1975The effect of a number of inhibitors of protein synthesis on ppGpp and pppGpp synthesis in vitro has been examined. As expected from in vivo results, chloramphenicol is without effect on this reaction. Aurintricarboxylic acid and chlortetracycline, on the other hand rapidly and specifically inhibit ppGpp synthesis.

Anomalous synthesis of ppGpp in growing cells
Cell, 1977In E. coli cells, accumulation of ppGpp is normally triggered by conditions that restrict the aminoacylation of tRNA or interfere with carbon/energy source metabolism; in both cases, the nucleotide's accumulation is associated with control of stable RNA synthesis and is generally believed to bring it about.

ppGpp: a global regulator in Escherichia coli
Trends in Microbiology, 2005The small nucleotide ppGpp acts as a global regulator of gene expression in bacteria. Proteomic analysis of cells lacking ppGpp has shown that this nucleotide might affect many more genes than previously anticipated. These findings and others suggest that ppGpp causes a redirection of transcription so that genes important for starvation survival and ...

Effect of ppGpp on the accuracy of protein biosynthesis
Archives of Biochemistry and Biophysics, 1983The maintenance of accuracy in protein biosynthesis in amino acid-starved rel+ strains of Escherichia coli has been attributed to an effect of ppGpp on the accuracy of aa-tRNA selection by the ribosome. It has been determined that concentrations of ppGpp characteristic of those found in amino acid-starved cells have no effect on the rate of reaction of

The magic dance of the alarmones (p)ppGpp
Molecular Microbiology, 2016SummaryThe alarmones (p)ppGpp are important second messengers that orchestrate pleiotropic adaptations of bacteria and plant chloroplasts in response to starvation and stress. Here, we review our structural and mechanistic knowledge on (p)ppGpp metabolism including their synthesis, degradation and interconversion by a highly diverse set of enzymes ...

Synthesis of ppGpp by mouse embryonic ribosomes
Cell, 1974Abstract The unusual nucleotide guanosine tetraphosphate (ppGpp) is synthesized in vitro by ribosomes isolated from mouse embryos, but is not formed by ribosomes isolated from adults. Analysis of specific organs shows a developmental change in detectable ppGpp-forming ability.
