Single‐Cell and Spatial Transcriptomic Profiling of Penile Squamous Cell Carcinoma Reveals Dynamics of Tumor Differentiation and Immune Microenvironment

open access: yesAdvanced Science, EarlyView.
This study uses single‐nucleus RNA sequencing and spatial transcriptomics to investigate penile squamous cell carcinoma (PSCC). It reveals that PSCC tumor cells mimic normal penile epithelium differentiation, independent of HPV status. The Tum_1 subtype shows basal stem‐like characteristics and promotes invasiveness. HPV‐positive basal stem‐like tumors

Organization of the Proteostasis Network of Membraneless Organelles

open access: yesAdvanced Science, EarlyView.
A comprehensive mapping of the proteostasis network (PN) across membraneless organelles (MLOs) reveals how central PN components coordinate MLO regulation. The analysis uncovers principles of specificity and promiscuity within the PN. The findings suggest that HSP70 chaperones act as generic MLO regulators, while client specific HSP70 co‐chaperones may

Analysis of in situ pre-mRNA targets of human splicing factor SF1 reveals a function in alternative splicing [PDF]

open access: yes, 2017
The conserved pre-mRNA splicing factor SF1 is implicated in 3′ splice site recognition by binding directly to the intron branch site. However, because SF1 is not essential for constitutive splicing, its role in pre-mRNA processing has remained mysterious.

Novel Antimicrobial Protein Fibroblast Growth Factor 8 Accelerates Skin Wound Healing via Directly Inhibiting Bacteria and Activating Glycolysis

open access: yesAdvanced Science, EarlyView.
Wound infections induce gcFGF8a expression, which subsequently executes direct antimicrobial activity to suppress local infection while simultaneously activating the FGFR4‐mediated ERK/AKT‐mTOR signaling cascade, thereby upregulating HIF1α and enhancing glycolysis. These coordinated actions synergistically promote tissue repair by eliminating pathogens

A Dual Approach with Organoid and CRISPR Screening Reveals ERCC6 as a Determinant of Cisplatin Resistance in Osteosarcoma

open access: yesAdvanced Science, EarlyView.
This study utilizes patient‐derived osteosarcoma organoids and CRISPR screening to identify ERCC6 and HNRNPM as key mediators of cisplatin resistance via the PI3K/AKT pathway and BAX alternative splicing. These findings provide novel insights and potential therapeutic targets for overcoming chemoresistance in osteosarcoma.

Regulated tissue-specific expression of antagonistic pre-mRNA splicing factors [PDF]

open access: yes, 1998
The SR proteins are essential metazoan pre-mRNA splicing factors that can also influence the selection of alternative 5' splice sites in a concentration-dependent manner.
