Ligand- and Structure-Based Pregnane X Receptor Models

2012
The human pregnane X receptor (PXR) is a ligand dependent transcription factor that can be activated by structurally diverse agonists including steroid hormones, bile acids, herbal drugs, and prescription medications. PXR regulates the transcription of several genes involved in xenobiotic detoxification and apoptosis.

Dexamethasone Induces Pregnane X Receptor and Retinoid X Receptor-α Expression in Human Hepatocytes: Synergistic Increase of CYP3A4 Induction by Pregnane X Receptor Activators

Molecular Pharmacology, 2000
In this report we show that submicromolar concentrations of dexamethasone enhance pregnane X receptor (PXR) activator-mediated CYP3A4 gene expression in cultured human hepatocytes. Because this result is only observed after 24 h of cotreatment and is inhibited by pretreatment with cycloheximide, we further investigated which factor(s), induced by ...

Pregnane X receptor: Predicting and preventing drug interactions

Thrombosis Research, 2005
The pregnane X receptor, PXR, is a member of the nuclear receptor family of ligand-activated transcription factors [1]. The hormones that act through the nuclear receptors include the steroid hormones, thyroid hormone, retinoic acid, and vitamin D [2].
