Rabbit prekallikrein

Rabbit prekallikrein (RPK) was purified from rabbit plasma by ion exchange and lectin column chromatography and preparative polyacrylamide gel electrophoresis. A 1500-fold purification was routinely achieved with a final yield of 5-10%. The purified RPK was found to be a glycoprotein with an apparent molecular weight of 88,000.

SUMMARY Two sibling miniature horses, a male and a female, had a normal 1-stage prothrombin time and a prolonged activated-partial thromboplastin time (aptt). The addition of as little as 5% of a normal equine plasma pool to the plasma samples of both horses shortened their prolonged aptt to within normal limits.

A method for the assay of human plasma prekallikrein in which a chromogenic synthetic tripeptide, PPAN, is used as a substrate for kallikrein is described. The conversion of prekallikrein to kallikrein is achieved by cold activation (0 degrees C) with water-soluble dextran sulfate.
