Longitudinal genome‐wide aneuploidy measurements in circulating cell‐free DNA to predict lack of benefit from pembrolizumab in patients with metastatic urothelial cancer

open access: yesMolecular Oncology, EarlyView.
Many patients with urothelial cancer do not benefit from treatment with pembrolizumab, while at risk of severe side effects. Changes in the levels of circulating tumor DNA early during treatment, measured by a simple and affordable assay that can be easily implemented in the clinic, can be used as a prognostic tool to identify these patients.

Deterministic and Probabilistic Assessment of Failure Mechanisms in Geosynthetic-Reinforced Embankments

open access: yesApplied Sciences
Geosynthetic-reinforced embankments are subject to two primary failure mechanisms: bond failure and rupture. Bond failure occurs when the critical slip surface extends beyond the reinforced zone, while rupture occurs when the slip surface intersects the ...

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

ZW4864‐mediated inhibition of the β‐catenin/BCL9/BCL9L complex reveals therapeutic potential in bladder cancer

open access: yesMolecular Oncology, EarlyView.
BCL9 and BCL9L drive bladder cancer progression by enhancing β‐catenin signaling, promoting proliferation, migration, invasion, and organoid growth. Genetic depletion of BCL9(L) suppresses malignant phenotypes, while pharmacological disruption of the β‐catenin/BCL9(L) complex with ZW4864 inhibits canonical Wnt signaling and tumor‐associated cellular ...

Random finite element analysis on ground subsidence caused by tunnel excavation in karst regions with spatial variable soil

open access: yesDeep Underground Science and Engineering
The construction of tunnels inevitably leads to ground subsidence, while surface collapses caused by caverns during excavation can result in significant financial losses.

Evaluation of the stability of a highway slope through numerical modeling

open access: yesDyna, 2017
Mass movement in natural slopes can cause great material and human loss for society. In this context, stability analyses become quite complex since there are a large number of uncertainties involved in the interpretation of the problem.

Single‐cell DNA methylation profiling: Technologies, computation, and applications in precision oncology

open access: yesMolecular Oncology, EarlyView.
Single‐cell DNA methylation (scDNAme) profiling maps epimutational clonal evolution, revealing mechanisms of malignancy and therapeutic resistance across diverse cancer types. By providing a high‐resolution landscape of intratumoral heterogeneity, these technologies empower precise patient stratification, guide the development of enhanced ...

Probabilistic Study on Subunit Mismatch Arrangement in Staphylococcal γ-hemolysin Heteroheptameric Transmembrane Pore

open access: yesJournal of Biomechanical Science and Engineering, 2011
Staphylococcal γ-hemolysin (Hlg), consists of two separate proteins, Hlg1 (LukF) of 34 kDa and Hlg2 of 32 kDa, which cooperatively lyse mammalian erythrocytes.

Interferon beta drives therapy resistance in a patient‐derived model of high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
Interferon type 1 (IFN‐1) production and signaling is associated with the acquisition of therapy resistance, following chronic DNA damage, via Interferon‐related DNA damage resistance signature (IRDS) gene expression. An alternative, DNA damage‐independent role of sustained IFN‐1 mediated resistance was identified and characterized by the emergence of ...
