Characterization of Human Prodynorphin Gene Transcripts

Prodynorphin gene transcripts have been characterized in human tissues with cRNA probes covering parts of the non-coding exon 1 and the main coding exon 4 using Northern blot and RNase protection experiments. A 2.8-kb signal was observed in human brain RNA with both the exon 1 and exon 4 probes.

The transcription factor cAMP response element binding (CREB) protein plays a pivotal role in drug-dependent neuronal plasticity. CREB phosphorylation at Ser133 is enhanced by drugs of abuse, including nicotine. Dynorphin (Dyn) contributes to the addictive process and its precursor gene prodynorphin (PD) is regulated by CREB.

Two regions of the human prodynorphin gene, the exon 4‐region coding for the opioid peptides and the putative promotor/exon 1‐region were analyzed for possible presence of polymorphisms. No polymorphism was detected in the exon 4‐region, whereas a GC/AT base‐pair exchange was observed 301 base pairs upstream of the exon 1/intron A boundary.

The regulation of prodynorphin gene expression by glucocorticoids in the hippocampus was examined in rats that were adrenalectomized (ADX) either 7, 30, 60 and 90 days prior to sacrifice. Peptide levels in the hippocampus of ADX rats were determined by radioimmunoassay and immunocytochemistry.
