PDGFA/PDGFRα signaling recruits CAFs to OSCC nests, and CAF‐OSCC contact drives tumor budding invasion. Mechanistically, heterotypic adhesion via E‐cadherin/integrin α2β1 activates YAP signaling in OSCC cells, inducing EMT. Concurrently, CAFs transfer mitochondria through TNTs, supplying ATP to fuel invasion.

Trypsinogen‐loaded nanoparticles reprogram TAMs via NF‐κB/NLRP3, driving M2→M1 conversion and potent phagocytosis to unleash antitumor immunity. ABSTRACT Although reprogramming tumor‐associated macrophages (TAMs) represents a promising therapeutic strategy, approaches that are both precise and safe remain scarce.

USP10 binds to and stabilizes PFKFB4, enhancing glycolytic ATP production, which activates the urea cycle and elevates fumarate. This inhibits histone demethylase KDM1A, leading to increased H3K4me1 enrichment at the Rad51 promoter and direct activation of Rad51 transcription, which confers lung cancer radioresistance. The PFKFB4 inhibitor 5MPN targets

LRRC4 suppresses GBM by regulating AP2A1‐containing Golgi‐derived clathrin‐coated vesicles. Low LRRC4 allows cytoplasmic AP2A1 to maintain mitochondrial fission‐fusion, MICOS integrity, and OXPHOS, promoting proliferation and invasion. High LRRC4 redirects AP2A1‐containing Golgi‐derived clathrin‐coated vesicles to mitochondria, disrupts MICOS, enhances

A double‐layered shell‐core microneedle patch is developed to co‐deliver FOLFIRINOX, surufatinib, and anti‐PD‐1 for localized chemo‐immunotherapy of PDAC. This strategy achieves sustained tumor suppression, reduces metastasis, and reprograms the TME by enhancing CD8+ T‐cell infiltration and inhibiting Tregs and M2 macrophages infiltration, while ...

Aberrant GALNT7‐mediated O‐GalNAcylation stabilizes TAZ to drive gallbladder cancer progression through a feed‐forward transcriptional loop. Structure‐based screening identifies Olaparib as a potent GALNT7 antagonist that disrupts this oncogenic axis, providing an immediate therapeutic strategy for this aggressive malignancy.

Protein Kinase R and the Inflammasome
Journal of Interferon & Cytokine Research, 2014Protein kinase R (PKR) was first identified as a mediator of the double-stranded RNA (dsRNA)–mediated inhibition of protein synthesis in extracts from interferon-treated cells. In a physiological context, viral replication results in production of dsRNA, activation of PKR by autophosphorylation, and phosphorylation of the protein synthesis initiation ...

The Role of Protein Kinase R in the Interferon Response
Journal of Interferon & Cytokine Research, 2011The effects of interferons (IFNs) are mediated through the induction of around 2,000 IFN-stimulated gene (ISG) products. However, the majority of these ISGs do not directly instigate IFN-mediated states, such as the defining resistance to viral infection.
