SPHINX31 acts as a SRPK1 inhibitor targeting the ATR/DNA‐PKcs/CHK1 replicative checkpoint to inhibit cell growth in non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
The kinase SRPK1 directly interacts with the protein TOPBP1 and regulates the pre‐mRNA splicing of WIZ thereby contributing to the activation of the ATR/CHK1 replicative checkpoint in response to replicative stress. This allows cancer cells' genomic stability and survival.

Functional characterisation of a predicted chloroplastic plant protein phosphatase [PDF]

open access: yes, 2009
The phosphatase AtPTPKISI is involved in the control of starch metabolism in Arabidopsis thaliana leaves at night. The SEX4 (Starch Excess 4) mutants, lacking this predicted phosphatase, have strongly reduced rates of starch metabolism.

How it all Started: Tau and Protein Phosphatase 2A

open access: yesJournal of Alzheimer's Disease, 2013
This review is dedicated to Inge Grundke-Iqbal who laid the foundations of the tau field, by isolating tau from the Alzheimer's disease (AD) brain, discovering that tau is hyperphosphorylated, and proving a critical role of protein phosphatase 2A (PP2A) and its endogenous inhibitor I2PP2A in this process. This memorial starts with a few personal notes,

Characterization of therapeutic antibodies targeting lymphatic and vascular resistance factor for the treatment of hypervascularized and highly metastatic cancers

open access: yesMolecular Oncology, EarlyView.
LVRF (lymphatic and vascular resistance factor) is a VEGFA splice variant upregulated in response to anti‐angiogenic therapy that fosters tumor adaptation through enhanced vascularization, lymphangiogenesis, and metastatic dissemination. Targeting LVRF with fully human neutralizing antibodies disrupts these processes and limits resistant tumor growth ...

Ginsenoside Rd attenuates beta-amyloid-induced tau phosphorylation by altering the functional balance of glycogen synthase kinase 3beta and protein phosphatase 2A

open access: yesNeurobiology of Disease, 2013
Neurofibrillary tangles are aggregates of hyperphosphorylated tau that are one of the pathological hallmarks of Alzheimer's disease (AD). Tau phosphorylation is regulated by a balance of kinase and phosphatase activities.

Optimization of seminal extracellular vesicle isolation protocols for cancer biomarker analysis using proteomic profiling

open access: yesMolecular Oncology, EarlyView.
This paper shows that size‐exclusion chromatography and sequential centrifugation applied to seminal plasma enrich for different populations of extracellular vesicles. This may significantly affect the detection and analysis of extracellular vesicle‐associated cancer biomarkers.

Identification of a novel protein phosphatase catalytic subunit by cDNA cloning

open access: yes, 1988
A cDNA encoding a novel protein phosphatase catalytic subunit (protein phosphatase X) has been isolated from a rabbit liver library. It codes for a protein having 45% and 65% amino acid sequence identity, respectively, to the catalytic subunits of ...

USP7 inhibits FOXO1 activity in chronic lymphocytic leukaemia cells enhancing cell survival

open access: yesMolecular Oncology, EarlyView.
CLL cells rely on BCR‐mediated signalling for survival and proliferation in part through suppression of FOXO1. FOXO1 activity is regulated by a plethora of post‐translational modifications including phosphorylation and ubiquitination. We demonstrated that dual inhibition of BCR signalling and the deubiquitinase USP7 activates FOXO1, modulating its ...

Distinct type-2A protein phosphatases activate HMGCoA reductase and acetyl-CoA carboxylase in liver

open access: yes, 1997
Acetyl-CoA carboxylase and HMGCoA reductase are inactivated by the same AMP-activated protein kinase and are activated by type-2A protein phosphatase.

Hyperosmotic stress‐induced redistribution of pre‐mRNA cleavage factor I subunits is associated with shifts in alternative polyadenylation

open access: yesFEBS Open Bio, EarlyView.
Hyperosmotic stress triggers the relocation of the CFIm complex from the nucleus to the cytoplasm. This shift creates a nuclear ‘stoichiometric bottleneck’, limiting CFIm availability for mRNA processing. Consequently, specific mRNAs like NUDT21 and DICER1 undergo targeted 3′UTR shortening, demonstrating how spatial protein dynamics drive rapid ...
