In the diagnosis of polycythaemia vera and essential thrombocythaemia, two molecular markers were described in the last decade: the overexpression of the PRV-1 gene and the V617F mutation in the JAK2 gene. In this study we assess their usefulness by comparing our test results with the available clinical data.

AbstractRecently, a Jak2V617F mutation has been described in the vast majority of patients with polycythemia vera (PV) as well as in subsets of patients with essential thrombocythemia (ET) and idiopathic myelofibrosis (IMF). The question arises whether this mutation is observed in those patients with ET and IMF who have also displayed previously ...

Abstract Polycythemia rubra vera-1 (PRV-1) is a member of the uPAR/CD59/Ly6 family of cell surface receptors. An increase in the expression level of PRV-1 mRNA has been shown in patients with polycythemia vera and essential thrombocythemia, however, the ligand and function of PRV-1 remains unclear.

Abstract Over the last years, PRV-1 expression has been considered to be closely connected with polycythemia vera (PV). However, the precise function of the PRV-1 protein has never been clarified while the search for the pathogenetic mechanism of PV has shifted to the association between the PRV-1 gene and the JAK2 V617F mutation rather ...

Clinical Utility of PRV-1 mRNA Expression Levels in Polycythemia Vera and Essential Thrombocythemia.
Blood, 2012Abstract Abstract 2846 Background: PRV-1 mRNA is overexpressed in most patients with polycythemia vera (PV) and a part of patients with essential thrombocythemia (ET). It is well known that PRV-1 expression levels correlate with JAK2 V617F mutation. While it was reported that JAK2 V617F

AbstractPolycythemia vera (PV) is a clonal stem cell disorder characterized by hyperproliferation of the erythroid, myeloid, and megakaryocytic lineages. Although it has been shown that progenitor cells of patients with PV are hypersensitive to several growth factors, the molecular pathogenesis of this disease remains unknown.

In a recent article in this journal, Klippel et al[1][1] reported that overexpression of the polycythemia rubra vera 1 gene (PRV-1) in purified granulocytes can distinguish patients with polycythemia vera (PV) from those with secondary erythrocytosis (SE) and from healthy subjects.

Abstract Background : Controversy persists regarding diagnostic accuracy as well as added value for quantitative assessment of neutrophil PRV-1 expression in polycythemia vera (Klippel et al. Blood2003;102:3569; Tefferi et al. Blood2004;103:3547).

Abstract Neutrophil PRV-1 mRNA is overexpressed in nearly all patients with polycythemia vera (PV) and discriminates PV from secondary polycythemia. In addition, 40– 50% of patients with essential thrombocythemia (ET) and chronic idiopathic myelofibrosis (cIMF) overexpress PRV-1.

Three consecutive polycythemia vera (PV) patients were analyzed before and during pegylated-interferon (rIFNalpha) treatment for the following markers: (1) granulocyte and CD34(+) cell clonality, (2) Jak2V617F expression, (3) PRV-1 mRNA overexpression, and (4) Epo-independent colony (EEC) growth.
