Role of Cysteine Residues in Pseudouridine Synthases of Different Families
Biochemistry, 1999The pseudouridine synthases catalyze the isomerization of uridine to pseudouridine in RNA molecules. An attractive mechanism was proposed based on that of thymidylate synthase, in which the thiol(ate) group of a cysteine side chain serves as the nucleophile in a Michael addition to C6 of the isomerized uridine.

tRNA Binding, Positioning, and Modification by the Pseudouridine Synthase Pus10
Journal of Molecular Biology, 2013Pus10 is the most recently identified pseudouridine synthase found in archaea and higher eukaryotes. It modifies uridine 55 in the TΨC arm of tRNAs. Here, we report the first quantitative biochemical analysis of tRNA binding and pseudouridine formation by Pyrococcus furiosus Pus10.

Molecular Recognition of tRNA by tRNA Pseudouridine 55 Synthase
Biochemistry, 1998Escherichia coli tRNA pseudouridine 55 synthase catalyzes pseudouridine formation at U55 in tRNA. A 17 base oligoribonucleotide analog of the T-arm was equivalent to intact native tRNA as a substrate for pseudouridine 55 synthase, viz., the features for substrate recognition by this enzyme are completely contained within the T-arm.

Pseudouridine Modifications in Transfer RNA and tRNA Pseudouridine Synthases
Journal of Molecular BiologyAmong the various modifications found in transfer RNAs, pseudouridine occurs the most frequently in all organisms and is also found in other RNA species including ribosomal, messenger, small nuclear, small nucleolar, and transfer-messenger RNA. Since the first gene encoding a tRNA pseudouridine synthase (truA) was discovered in 1978, many pseudouridine

Spatially controlled expression of the Drosophila pseudouridine synthase RluA-1
The International Journal of Developmental Biology, 2011Pseudouridine (Ψ) synthases function in the formation of Ψ, the most abundant of the modified RNA residues. All Ψ synthases in E. coli are classified into one of five families according to their sequences. Among them, members of the RluA Ψ synthase family catalyze certain Ψ formations in ribosomal RNA.

The structural basis for tRNA recognition and pseudouridine formation by pseudouridine synthase I.
Nature structural biology, 2000Pseudouridine synthases catalyze the isomerization of specific uridines to pseudouridine in a variety of RNAs, yet the basis for recognition of the RNA sites or how they catalyze this reaction is unknown. The crystal structure of pseudouridine synthase I from Escherichia coli, which, for example, modifies positions 38, 39 and/or 40 in tRNA, reveals a ...
