The interactions between biogenic amines and pyridoxal, pyridoxal phosphate, and pyridoxal kinase

Archives of Biochemistry and Biophysics, 1972
Abstract The effect of biogenic amines on pyridoxal kinase activity has been investigated in vitro . Experiments with a purified beef brain pyridoxal kinase indicate that 3,4-dihydroxyphenylalanine (dopa), dopamine, norepinephrine, and serotonin inhibit the reaction, while tyrosine, tryptophan, and 5-hydroxytryptophan (5-HTP) have no effect. However,

Fluorometric assay of pyridoxal and pyridoxal 5′-phosphate

Analytical Biochemistry, 1979
Abstract A new and very sensitive fluorometric method for the determination of pyridoxal and pyridoxal 5′-phosphate is reported. The specificity is based on the reductive amination of pyridoxal and its 5′-phosphate with methyl anthranilate and sodium cyanoborohydride at pH 4,5 to 5,0.

Binding of a photoaffinity analogue of pyridoxal to pyridoxal kinase

European Journal of Biochemistry, 1990
The binding of pyridoxal analogues to the structural domains of pyridoxal kinase was studied by fluorescence spectroscopy and chromatographic techniques. Two fragments of 24 and 16 kDa, arising from limited proteolysis of the native enzyme, were separated by ion‐exchange chromatography and used for binding studies with pyridoxal oxime.

Inhibition of gaba binding by pyridoxal and pyridoxal phosphate

International Journal of Biochemistry, 1980
Abstract 1. 1. GABA exerts its neuroinhibitory effects by binding to a specific receptor on the post-synaptic membrane. 2. 2. Since both GABA and vitamin B 6 have been implicated in convulsion, the effects of B 6 vitamers on Na-independent binding of GABA was investigated by incubating synaptic membrane suspensions for 5 min at 4°C in 1 ml ...

Measurement of pyridoxal 5′-phosphate, pyridoxal, and 4-pyridoxic acid in the cerebrospinal fluid of children

Clinica Chimica Acta, 2017
We quantified pyridoxal 5'-phosphate (PLP), pyridoxal (PL), and 4-pyridoxic acid (PA) in the cerebrospinal fluid (CSF) of children and to investigate the effect of age, sex, epilepsy, and anti-epileptic drug (AED) therapy on these vitamers.CSF samples prospectively collected from 116 pediatric patients were analyzed. PLP, PL, and PA were measured using

New Pathway of Conversion of Pyridoxal to 4-Pyridoxic Acid

Enzyme, 2017
The only enzyme demonstrated so far to catalyze the formation of 4-pyridoxic acid, the final excretory product of vitamin B(6), was aldehyde oxidase. In this paper we have presented an evidence that another enzyme, NAD^+-dependent aldehyde dehydrogenase, is capable of catalyzing this reaction. Rat mutants with high, low and no aldehyde oxidase activity

The triplet state of pyridoxal

Biochemical and Biophysical Research Communications, 1968
Abstract The absorption spectra, as well as the relative distribution of the structures of pyridoxal in aqueous solution, have been investigated in several laboratories ( Williams and Neilands, 1954 ; Metzler and Snell, 1955 ; Anderson and Martell, 1964) .

Pyridoxal 5′-phosphate, pyridoxal, and 4-pyridoxic acid in the paired serum and cerebrospinal fluid of children

Clinica Chimica Acta, 2017
We quantified pyridoxal 5'-phosphate (PLP), pyridoxal (PL), and 4-pyridoxic acid (PA) in paired serum and cerebrospinal fluid (CSF) samples from children and investigated the effect of age on the concentrations and CSF-to-serum ratios of these vitamers.Serum and CSF samples prospectively collected from 49 pediatric patients were analyzed.

Pyridoxal Kinase

Annals of the New York Academy of Sciences, 1990
Chymotryptic digestion of sheep brain pyridoxal kinase, a dimer of identical subunits each of 40 kDa, yields two fragments of 24 and 16 kDa with concomitant loss of catalytic activity. These fragments were separated by HPLC and used for binding studies with ATP and pyridoxal analogues.

Pyridoxal-5′-phosphate and pyridoxal biokinetics in aging Wistar rats

Experimental Gerontology, 1991
Biokinetic parameters of plasma pyridoxal-5'-phosphate (PLP) and pyridoxal (PL) disposition were studied in male Wistar rats aged 8 and 27 months kept from weaning on a purified diet containing 250 g casein and 6 mg pyridoxine.HCl per kg. Baseline plasma PLP concentration was lower in the older animals (514 +/- 56 nmol/L for young and 317 +/- 124 nmol ...
