Syntheses and reactions of thieno [2,3-d] pyrimidines [PDF]
Thieno [2,3-d] pyrimidines have, so far, been synthesised mainly from thiophen derivatives. The work described in this thesis is largely concerned with a search for general methods of synthesising substituted thieno [2,3-d] pyrimidines from ...

The first enantioselective alkynylations of aqueous fluoral are described. These processes are catalyzed by an iodide‐bound ruthenium‐(Ph‐BPE)‐catalyst; an octahedral metal complex that can exist as 12 diastereomeric‐at‐metal isomers. Calculations reveal that 2 of 12 stereoisomeric complexes account for 99.9% of the Boltzmann population and that ...

Modular synthesis of the pyrimidine core of the manzacidins by divergent Tsuji–Trost coupling
The design, development and application of an efficient procedure for the concise synthesis of the 1,3-syn- and anti-tetrahydropyrimidine cores of manzacidins are reported.

Objective Methotrexate (MTX) is the first‐line therapy for juvenile idiopathic arthritis (JIA), but up to 40% of patients do not respond to it. Low inosine triphosphate pyrophosphatase (ITPA) activity has been associated with reduced clinical remission. We investigated the role and underlying mechanisms of ITPA in vitro. Methods ITPA enzymatic activity

A Modular Nucleoside Kinase Cascade for the Synthesis of Ribonucleoside Triphosphates
Damm et al. report standardized enzymatic cascades for production of all canonical nucleoside triphosphates from the corresponding nucleosides, using kinase‐catalyzed phosphorylation from acetyl phosphate. Reactions afforded ∼ 100 mM product in near‐quantitative conversion.

Targeting HPK1 is a promising strategy to restore T‐cell anti‐tumor immunity. We report the design and synthesis of novel 2,4‐diaminopyrimidine derivatives, focusing on systematic structure–activity relationship (SAR) analysis These findings identify key structural features for potent HPK1 inhibition, facilitating the discovery of next‐generation ...

Pyrrolo[2,3-d]pyrimidines : design, synthesis, functionalisation

Solid-phase synthesis of tetrasubstituted pyrrolo[2,3-d]pyrimidines
A facile solid phase synthesis of 2,4,6,7-tetrasubstituted pyrrolo[2,3-d]pyrimidines is described. The synthesis involves a highly efficient five-step route starting from resin-bound dimeric peptoids.
