Analysis of the catalytic mechanism of pyruvate dehydrogenase kinase

Archives of Biochemistry and Biophysics, 2005
It has been proposed that "Glu238" within the N-box of pyruvate dehydrogenase kinase (PDK) is a base catalyst. The pH dependence of k(cat) of Arabidopsis thaliana PDK indicates that ionizable groups with pK values of 6.2 and 8.4 are necessary for catalysis, and the temperature dependence of these values suggests that the acidic pK is due to a carboxyl-

Specificity of the pyruvate dehydrogenase kinase for pyruvate dehydrogenase component bound to the surface of the kidney pyruvate dehydrogenase complex and evidence for intracore migration of pyruvate dehydrogenase component

Biochemistry, 1983
Using the bovine kidney pyruvate dehydrogenase complex we have investigated the mechanism whereby about three pyruvate dehydrogenase (active form) kinase molecules, tightly bound to the dihydrolipoyl transacetylase core, can rapidly phosphorylate and inactivate about 20 pyruvate dehydrogenase (active form) (PDHa) tetramers which are also bound to the ...

Mechanism of pyruvate inhibition of plant pyruvate dehydrogenase kinase and synergism with ADP

Archives of Biochemistry and Biophysics, 1990
The effects of various metabolites on pyruvate dehydrogenase (PDH) kinase-catalyzed inactivation of the pyruvate dehydrogenase complex (PDC) were studied in extracts of mitochondria purified from green leaf tissue of Pisum sativum L. Pyruvate was an uncompetitive inhibitor of PDH kinase with respect to ATP whereas ADP was a competitive inhibitor.

AZD7545 is a selective inhibitor of pyruvate dehydrogenase kinase 2

Biochemical Society Transactions, 2003
The PDH (pyruvate dehydrogenase) multi-enzyme complex catalyses a key regulatory step in oxidative glycolysis. Phosphorylation of the E1 subunit of the complex on serine residues results in the inactivation of enzyme activity. A family of four dedicated PDH kinase isoenzymes exists, each of which displays a distinct tissue-specific expression profile ...

Triterpene and diterpene inhibitors of pyruvate dehydrogenase kinase (PDK)

Bioorganic & Medicinal Chemistry Letters, 1999
Several oximes of triterpenes with a 17-beta hydroxyl and abietane derivatives are inhibitors of pyruvate dehydrogenase kinase (PDK) activity. The oxime 12 and dehydroabietyl amine 2 exhibit a blood glucose lowering effect in the diabetic ob/ob mouse after a single oral dose of 100 micromol/kg.

The immunometabolic role of pyruvate dehydrogenase kinase/pyruvate dehydrogenase axis in cardiovascular disease and beyond

Cardiovascular Research
Abstract The pyruvate dehydrogenase kinase (PDK)/pyruvate dehydrogenase (PDH) axis plays a pivotal role in regulating cellular energy metabolism, controlling the flux between the tricarboxylic acid cycle and oxidative phosphorylation.
