We show that the majority of the 18 analyzed recurrent cancer‐associated ERBB4 mutations are transforming. The most potent mutations are activating, co‐operate with other ERBB receptors, and are sensitive to pan‐ERBB inhibitors. Activating ERBB4 mutations also promote therapy resistance in EGFR‐mutant lung cancer.

Methods to improve antibody–drug conjugate (ADC) treatment durability in cancer therapy are needed. We utilized ADCs and immune‐stimulating antibody conjugates (ISACs), which are made from two non‐competitive antibodies, to enhance the entry of toxic payloads into cancer cells and deliver immunostimulatory agents into immune cells.

Interrupted quenching in high carbon steels for forgings
The present study is focused on analyzing the effect of the interrupted quenching followed by a partitioning process in a high carbon steel 0,50 % C, 1,50 % Mn, 0,40 % Si, 2,00 % Cr without significant contribution of Al.

RIPK4 function interferes with melanoma cell adhesion and metastasis
RIPK4 promotes melanoma growth and spread. RIPK4 levels increase as skin lesions progress to melanoma. CRISPR/Cas9‐mediated deletion of RIPK4 causes melanoma cells to form less compact spheroids, reduces their migratory and invasive abilities and limits tumour growth and dissemination in mouse models.

Homologous expression and purification of human HAX‐1 for structural studies
This research protocol provides detailed instructions for cloning, expressing, and purifying large quantities of the intrinsically disordered human HAX‐1 protein, N‐terminally fused to a cleavable superfolder GFP, from mammalian cells. HAX‐1 is predicted to undergo posttranslational modifications and to interact with membranes, various cellular ...

Light activation of the photoswitchable cannabinoid ligand azo‐HU308 triggers Ca2+ influx in pancreatic β‐cells through TRPC channels, independent of CB2 cannabinoid receptors. This reveals a non‐GPCR pathway for cannabinoid modulation of β‐cell Ca2+ dynamics and establishes azo‐HU308 as an optical tool to study cannabinoid signaling through TRP ...

Error Propagation Analysis for Quantitative Intracellular Metabolomics
Model-based analyses have become an integral part of modern metabolic engineering and systems biology in order to gain knowledge about complex and not directly observable cellular processes. For quantitative analyses, not only experimental data, but also

Inhomogeneous thermal quenches [PDF]
We describe holographic thermal quenches that are inhomogeneous in space. The main characteristic of the quench is to take the system far from its equilibrium configuration. Except special extreme cases, the problem has no analytic solution. Using the numerical holography methods, we study different observables that measure thermalization such as the ...
