Higher Rates of Hypersensitivity Reactions to Calaspargase Compared With Pegaspargase: A Single Center Retrospective Review

open access: yesPediatric Blood &Cancer, Volume 72, Issue 5, May 2025.
ABSTRACT Background Asparaginase is a critical component of curative therapy for the treatment of pediatric acute lymphoblastic leukemia (ALL) and acute lymphoblastic lymphoma (LLy). Pegaspargase (PEG) increased the duration of asparagine depletion and lowered the incidence of hypersensitivity reactions (HSRs) compared with native asparaginase.

Drug Interactions With Tamoxifen and Treatment Effectiveness in Premenopausal Breast Cancer Patients: A Bayesian Joint Modeling Approach

open access: yesPharmacoepidemiology and Drug Safety, Volume 34, Issue 5, May 2025.
ABSTRACT Purpose Tamoxifen is guideline treatment for premenopausal women with estrogen receptor‐positive (ER+) breast cancer. Therapeutic efficacy relies partly on tamoxifen biotransformation by CYP2D6, CYP2C19, and CYP3A4 enzymes. We conducted a cohort study to evaluate whether concomitant prescription of drugs that inhibit these enzymes impacted ...

An Enhanced Metabolization Protocol for In Vitro Genotoxicity Assessment of N‐Nitrosamines in Mammalian Cells

open access: yesEnvironmental and Molecular Mutagenesis, Volume 66, Issue 4, Page 210-220, April 2025.
ABSTRACT N‐Nitrosamines (NAs) are probable human carcinogens and were detected as impurities in pharmaceuticals, which led to a concern for human health. NAs require metabolic activation before they become mutagenic, and not all NAs are mutagenic since their reactivity is related to their structure.
