Reversing Macrophage Immunometabolic Dysfunction in Prolonged Infections Via a Glutaminolysis‐Fueled Restoration‐Based Hierarchically Selective Therapeutic System

open access: yesAdvanced Science, EarlyView.
A hierarchically selective immunotherapeutic nanoplatform, mGls@HEV‐MTP, selectively restores macrophage immunocompetence through GLS1‐mediated restoration of glutaminolysis‐fueled anaplerosis. Concurrently, it renders Staphylococcus aureus (S. aureus) more readily recognizable to the metabolically reprogrammed macrophages, thereby facilitating ...

Cadmium activates NLRP3 inflammasome-mediated pyroptosis in murine macrophage cell line RAW 264.7

open access: yes, 2022
Introduction: Cadmium (Cd) is an environmental toxin that affects the kidney, liver and lungs. Its effect on innate immunity is not well known. Methods: RAW 264.7 macrophage cells were treated with different concentrations of CdCl₂ (5, 10, 15 μM) for ...

Quantitative Visualization of Intracellular Nanometabolism Using Peak‐Shifted Dual‐State Emissive FRET Nanoprobes

open access: yesAdvanced Science, EarlyView.
This study reveals the long‐elusive intracellular dissolution mechanism of carrier‐free nanomedicines. By utilizing peak‐shifted dual‐state emissive FRET nanoprobes (PDFNPs) that undergo a distinct ratiometric fluorescence peak shift upon disassembly, we achieve real‐time, quantitative tracking of dissolution kinetics in live cells.

lncRNA NEAT1 mediates sepsis progression by regulating Irak2 via sponging miR-370-3p

open access: yesBiology Open, 2020
Sepsis is a life-threatening condition and often associated with multiple organ failure. Nuclear-enriched abundant transcript 1 (NEAT1), a member of the long non-coding RNAs (lncRNAs), was reported to be involved in the regulation of sepsis progression ...

Aldosterone induced galectin-3 expression mainly through MRs in macrophage cell line Raw 264.7.

open access: yes, 2014
Raw 264.7 cells(1×106/ml) were serum-starved for 24 h, then treated with 10−7 M of spironolactone or 10−7 M of RU-486 for 1 h, followed by 10−6 M aldosterone or vehicle treatment for 24 h.

An Injectable ROS‐Responsive Nanozyme Hydrogel Regulates the Uterine Microenvironment to Prevent Intrauterine Adhesions

open access: yesAdvanced Science, EarlyView.
The injectable CeTA@GPP hydrogel is administered via intrauterine injection, forming a protective barrier. In the high ROS inflammatory microenvironment of injured endometrium, boronate ester cleavage triggers responsive CeTA release and local enrichment.

RAW 264.7 cell phagocytosis assay v1

open access: yes
This protocol describes in vitro methods for evaluating phagocytosis using the macrophage-like cell line, RAW 264.7. The two methods discussed here include (1) a photographic phagocytosis assay, which is compatible with any target that is visible via 100x microscopy, and measures total uptake of target material per macrophage, and (2) the phagocytosis ...

Force‐Electric Nanotentacles of Tantalum Implant Accelerate Cellular Energy Metabolism/Crosstalk for Immunomodulation‐Vascularized Bone Integration

open access: yesAdvanced Science, EarlyView.
A biomimetic force‐electric nanotentacle‐enabled tantalum (Ta) implant with surface‐nanostructured LiTaO3 (n‐LiTaO3) was first developed via laser nanofabrication combined with Li+‐induced in situ reaction, enabling the transduction of ultrasound into bioelectrical cues.

Berberine Inhibits the Inflammatory Response Induced by Staphylococcus aureus Isolated from Atopic Eczema Patients via the TNF-α/Inflammation/RAGE Pathways

open access: yesCells
Atopic eczema patients exhibit high levels of Staphylococcus aureus (S. aureus) skin colonization. S. aureus can stimulate macrophages and the expression of proinflammatory cytokines. Berberine (BBR), an alkaloid, attenuates S.
