A Twenty-First Century Cancer Epidemic Caused by Obesity: The Involvement of Insulin, Diabetes, and Insulin-Like Growth Factors

open access: yesInternational Journal of Endocrinology, 2013
Obesity has reached epidemic proportions in the developed world. The progression from obesity to diabetes mellitus type 2, via metabolic syndrome, is recognised, and the significant associated increase in the risk of major human cancers acknowledged.

Decoding Human Placental Cellular and Molecular Responses to Obesity and Fetal Growth

open access: yesAdvanced Science, EarlyView.
Women with obesity often deliver large‐for‐gestational‐age (LGA) infants. Single‐nucleus RNA sequencing of term placenta reveals that hypoxia and TNF‐α signaling in syncytiotrophoblasts are featured in maternal obesity, but inflammatory signatures in Hofbauer cells and response to lipid or carbohydrate metabolism in fibroblasts are specific to LGA.

Correlations between IGF-IR Expression and Clinicopathological Factors and Prognosis in Patients with Lung Adenocarcinoma

open access: yesChinese Journal of Lung Cancer, 2010
Background and objective The incidence of lung adenocarcinoma increases rapidly, and IGF-IR is the key mediator of several growth factors signal transduction, therefore it plays an important role in the proliferation and differentiation of cancer cell ...

The tobacco-specific carcinogen NNK induces pulmonary tumorigenesis via nAChR/Src/STAT3-mediated activation of the renin-angiotensin system and IGF-1R signaling

open access: yesExperimental and Molecular Medicine, 2023
The renin-angiotensin (RA) system has been implicated in lung tumorigenesis without detailed mechanistic elucidation. Here, we demonstrate that exposure to the representative tobacco-specific carcinogen nitrosamine 4-(methylnitrosamino)-1-(3-pyridyl)-1 ...

Dual silencing of the EGF and type 1 IGF receptors suggests dominance of IGF signaling in human breast cancer cells

open access: yesBiochemical and Biophysical Research Communications, 2007
Signaling via the type 1 insulin-like growth factor receptor (IGF1R) confers resistance to EGF receptor (EGFR) inhibitors. It is plausible that reciprocal EGFR compensation could mediate resistance to IGF1R inhibition, prompting us to investigate effects of IGF1R depletion on EGFR signaling in breast cancer cells expressing relatively high (MDA-MB-468)

RUNX2 Activation in Fibro/Adipogenic Progenitors Promotes Muscle Fibrosis in Muscular Dystrophy

open access: yesAdvanced Science, EarlyView.
This study revealed a novel role of the chemokine‐TGF‐β1‐RUNX2 axis in determining the fate of FAP differentiation and modulating muscle fibrosis in patients and mice with muscular dystrophies. ABSTRACT Clinical evidence indicates concurrent muscle inflammation and fibrosis in muscular dystrophies (MDs); however, the molecular mechanisms underlying ...

Targeting the Insulin-Like Growth Factor (IGF) System Is Not as Simple as Just Targeting the Type 1 IGF Receptor

open access: yesAmerican Society of Clinical Oncology Educational Book, 2012
Overview: Increased signaling of the insulin-like growth factor (IGF) system via alterations in expression levels of its components has been demonstrated in various tumor types. Numerous experimental studies have supported the involvement of the IGF system signaling axis in tumor initiation and progression.

Conformable Device for Independent Measurements of Mucosal and Vascular Barriers in a Complex In Vitro Intestinal Model

open access: yesAdvanced Science, EarlyView.
Development of more complex in vitro models necessitates the development of improved monitoring approaches. This work demonstrates a novel conformable device capable of measuring the resistance of two cell barriers within a single model independently, a significant advantage over conventional trans‐epithelial/endothelial electrical resistance (TEER ...

Engineered GM1 Intersects Between Mitochondrial and Synaptic Pathways to Ameliorate ALS Pathology

open access: yesAdvanced Science, EarlyView.
Amyotrophic Lateral Sclerosis (ALS) is a fatal neurodegenerative disease driven by genetic and molecular disruptions affecting energy balance, protein homeostasis, and stress responses in nerve cells. Studies using human and rodent models identified convergent defects in mitochondria and synaptic function.

Amniotic IGF-I supplementation of growth-restricted fetal sheep alters IGF-I and IGF receptor type 1 mRNA and protein levels in placental and fetal tissues [PDF]

open access: yesJournal of Endocrinology, 2005
We have previously reported that chronic intra-amniotic supplementation of the late gestation growth-restricted (IUGR) ovine fetus with IGF-I (20 μg/day) increased gut growth but reduced liver weight and circulating IGF-I concentrations. Here we report mRNA and protein levels of IGF-I, the type 1 IGF receptor (IGF-1R) and IGF-binding proteins (IGFBP)-1,
