Melanocortin Receptors: Ligands and Proteochemometrics Modeling
Annals of the New York Academy of Sciences, 2003Abstract: The melanocortin receptors exist in five subtypes, MC1–5R. These receptors participate in important regulations of the immune system, central behavior, and endocrine and exocrine glands. Here we provide a short review on MCR subtype selective peptides and organic compounds with activity on the MCRs, developed in our laboratory. Also provided

Melanocortin-4 receptor mutations in obesity.
Advances in clinical chemistry, 2009The current alarming spread of obesity in many parts of the world is caused by a sudden environmental shift characterized by replacement of a frugal diet with low cost, energy dense food, and little requests for physical activity during work and leisure time.

Although human pigmentation is genetically complex, to date polymorphism at only 1 locus, the melanocortin-1 receptor (MC1-R), has been associated with physiologic variation in hair and skin color. The MC1-R, a G protein-coupled receptor with 7 transmembrane-spanning domains, plays a key role in determining the type of melanin (eumelanin vs pheomelanin)

Melanocortins in Brain Inflammation: The Role of Melanocortin Receptor Subtypes
2010The melanocortins (MC) are released from neurons and paracrine cells in the CNS where they are involved in important physiological functions, including regulation of body temperature and immune responses. MC bind to melanocortin receptors, a class of cell surface G-protein-coupled receptors. Of the five subtypes of MC receptors that have been cloned in

Chapter 12 Melanocortin receptors
1997Publisher Summary Pro-opiomelanocortin (POMC) is expressed in the brain as well as the pituitary and is the precursor for melanocortin peptides, adrenocorticotropin, melanocyte-stimulating hormone, and fragments thereof. In addition to their well-known effects on pigmentation and steroidogenesis, melanocortin peptides have numerous activities in the ...

The melanocyte-stimulating hormone (MSH) receptor, recently renamed the melanocortin-1 receptor (MC1-R), is a 7 transmembrane domain receptor in the rhodopsin superfamily that plays an important role in the regulation of mammalian pigmentation. The study of the MC 1-R has introduced at least two novel paradigms to the G protein signaling field ...

Melanocortin receptors: perspectives for novel drugs
European Journal of Pharmacology, 1999The cloning of five different subtypes of melanocortin receptor subtypes have recently opened up new possibilities for the development of drugs. The physiological roles of the five melanocortin receptors have started to become understood, and compounds with selective actions on some of the five subtypes have become available. Presently, most clinically

After cloning of the melanocyte MC1-R (1,2) and adrenocortical MC2-R (2),interest in the possibility of unique neural homologs of these receptors grew from observations of central effects of melanocortins, such as effects on learning and memory (reviewed in ref. 3) and temperature control (4).

Melanocortins are a subset of peptides derived from the proopiomelanocortin (POMC) gene product, including ACTH, α-, β-, and γ-MSH (1,2). The name refers to the two principal activities of these peptides, that is, regulation of pigmentation in skin and hair and steroidogenesis in the adrenal cortex.

This chapter deals with perhaps the least understood receptor for the melanocortin peptides, that is the melanocortin-3 receptor (MC3-R). Although naturally occurring and genetically engineered mutations have provided us with insight into the function of the other known melanocortin receptors, little is known about the physiologic role of the MC3-R ...
