Generation of NSE-MerCreMer transgenic mice with tamoxifen inducible Cre activity in neurons. [PDF]

open access: yesPLoS ONE, 2012
To establish a genetic tool for conditional deletion or expression of gene in neurons in a temporally controlled manner, we generated a transgenic mouse (NSE-MerCreMer), which expressed a tamoxifen inducible type of Cre recombinase specifically in ...

Recombinase-based genetic circuit optimization

open access: yes2017 IEEE Biomedical Circuits and Systems Conference (BioCAS), 2017
The rapid advancements of synthetic biology show promising potential in biomedical and other applications. Recently, recombinases were proposed as a tool to engineer genetic logic circuits with long-term memory in living and even mammalian cells.

Spontaneous self-segregation of Rad51 and Dmc1 DNA recombinases within mixed recombinase filaments [PDF]

open access: yesJournal of Biological Chemistry, 2018
During meiosis, the two DNA recombinases Rad51 and Dmc1 form specialized presynaptic filaments that are adapted for performing recombination between homologous chromosomes. There is currently a limited understanding of how these two recombinases are organized within the meiotic presynaptic filament.

Aggressive prostate cancer is associated with pericyte dysfunction

open access: yesMolecular Oncology, EarlyView.
Tumor‐produced TGF‐β drives pericyte dysfunction in prostate cancer. This dysfunction is characterized by downregulation of some canonical pericyte markers (i.e., DES, CSPG4, and ACTA2) while maintaining the expression of others (i.e., PDGFRB, NOTCH3, and RGS5).

Tyrosine Recombinase Retrotransposons and Transposons

open access: yesMicrobiology Spectrum, 2015
ABSTRACT Retrotransposons carrying tyrosine recombinases (YR) are widespread in eukaryotes. The first described tyrosine recombinase mobile element, DIRS1, is a retroelement from the slime mold Dictyostelium discoideum . The YR elements are bordered by terminal repeats related to their replication via free

Transcriptional network analysis of PTEN‐protein‐deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication

open access: yesMolecular Oncology, EarlyView.
Combining PTEN protein assessment and transcriptomic profiling of prostate tumors, we uncovered a network enriched in senescence and extracellular matrix (ECM) programs associated with PTEN loss and conserved in a mouse model. We show that PTEN‐deficient cells trigger paracrine remodeling of the surrounding stroma and this information could help ...

The Bxb1 recombination system demonstrates heritable transmission of site-specific excision in Arabidopsis

open access: yesBMC Biotechnology, 2012
Background The mycobacteriophage large serine recombinase Bxb1 catalyzes site-specific recombination between its corresponding attP and attB recognition sites.

Computational characterization of recombinase circuits for periodic behaviors

open access: yesiScience, 2023
Summary: Recombinases are site-specific proteins found in nature that are capable of rearranging DNA. This function has made them promising gene editing tools in synthetic biology, as well as key elements in complex artificial gene circuits implementing ...

Characterization of an archaeal recombinase paralog that exhibits novel anti-recombinase activity

open access: yesMutation Research - Fundamental and Molecular Mechanisms of Mutagenesis, 2020
The process of homologous recombination is heavily dependent on the RecA family of recombinases for repair of DNA double-strand breaks. These recombinases are responsible for identifying homologies and forming heteroduplex DNA between substrate ssDNA and dsDNA templates, activities that are modified by various accessory factors.

Identification of serum protein biomarkers for pre‐cancerous lesions associated with pancreatic ductal adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This work identified serum proteins associated with pancreatic epithelial neoplasms (PanINs) and early‐stage PDAC. Proteomics screens assessed genetically engineered mice with abundant PanINs, KPC mice (Lox‐STOP‐Lox‐KrasG12D/+ Lox‐STOP‐Lox‐Trp53R172H/+ Pdx1‐Cre) before PDAC development and also early‐stage PDAC patients (n = 31), compared to benign ...
