To help end the replication crisis and instill confidence in our scientific literature, we introduce a new process for evaluating scientific manuscripts, termed “peer replication,” in which referees independently reproduce key experiments of a manuscript.

Evolutionary interplay between viruses and R‐loops
Viruses interact with specialized nucleic acid structures called R‐loops to influence host transcription, epigenetic states, latency, and immune evasion. This Perspective examines the roles of R‐loops in viral replication, integration, and silencing, and how viruses co‐opt or avoid these structures.

Structural Insights into APOBEC3-Mediated Lentiviral Restriction
Mammals have developed clever adaptive and innate immune defense mechanisms to protect against invading bacterial and viral pathogens. Human innate immunity is continuously evolving to expand the repertoire of restriction factors and one such family of ...

The Contribution of Lysosomes to DNA Replication
Lysosomes, acidic, membrane-bound organelles, are not only the core of the cellular recycling machinery, but they also serve as signaling hubs regulating various metabolic pathways. Lysosomes maintain energy homeostasis and provide pivotal substrates for

The authors of this paper were interested to see whether the expression of three proteins (A, B and C) was altered in a knockout mouse model of a gene encoding Protein X. Each experiment measured the expression of a different protein and the results seem to show a clear increase in protein expression in the mutant mouse compared with the control.

Disruption of SETD3‐mediated histidine‐73 methylation by the BWCFF‐associated β‐actin G74S mutation
The β‐actin G74S mutation causes altered interaction of actin with SETD3, reducing histidine‐73 methylation efficiency and forming two distinct actin variants. The variable ratio of these variants across cell types and developmental stages contributes to tissue‐specific phenotypical changes. This imbalance may impair actin dynamics and mechanosensitive

This study reveals how prime editing guide RNA (pegRNA) secondary structure and reverse transcriptase template length affect prime editing efficiency in correcting the phospholamban R14del cardiomyopathy‐associated mutation. Insights support the design of structurally optimized enhanced pegRNAs for precise gene therapy.
