Familial Medullary Thyroid Cancer: Five-year Review of the Most Frequent Mutations in the RET Gene: An Update

open access: yesSudan Journal of Medical Sciences, 2022
Background: Familial Medullary Thyroid Cancer (FMTC) is hereditary in 25% of cases. Patients with an inherited form of FMTC usually have a germline mutation in the RET proto-oncogene (10q11.2); these mutations generally occur in exons 10 (codons 618 and ...

Advances in the Diagnosis and Treatment of a Driving Target: RET Rearrangements in non-Small-Cell Lung Cancer (NSCLC) Especially in China

open access: yesTechnology in Cancer Research & Treatment, 2023
In the era of precision medicine, with the deepening of the research on malignant tumor driving genes, clinical oncology has fully entered the era of targeted therapy.

RET codon 618 mutations in Saudi families with multiple endocrine neoplasia Type 2A and familial medullary thyroid carcinoma

open access: yesAnnals of Saudi Medicine, 2013
BACKGROUND AND OBJECTIVES: Certain diseases such as multiple endocrine neoplasia (MEN) 2A, MEN 2B, familial and sporadic medullary thyroid carcinoma (MTC) and renal dysgenesis are related to abnormalities of the RET protein.

PTPRA Phosphatase Regulates GDNF-Dependent RET Signaling and Inhibits the RET Mutant MEN2A Oncogenic Potential

open access: yesiScience, 2020
Summary: The RET proto-oncogene encodes receptor tyrosine kinase, expressed primarily in tissues of neural crest origin. De-regulation of RET signaling is implicated in several human cancers.

Proteomic Identification of DNA-PK Involvement within the RET Signaling Pathway. [PDF]

open access: yesPLoS ONE, 2015
Constitutive activation of the Rearranged during Transfection (RET) proto-oncogene leads to the development of MEN2A medullary thyroid cancer (MTC). The relatively clear genotype/phenotype relationship seen with RET mutations and the development of MEN2A

Molecular mechanisms of RET receptor-mediated oncogenesis in multiple endocrine neoplasia 2

open access: yesClinics, 2012
Multiple endocrine neoplasia type 2 is an inherited cancer syndrome characterized by tumors of thyroid and adrenal tissues. Germline mutations of the REarranged during Transfection (RET) proto-oncogene, leading to its unregulated activation, are the ...

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Pralsetinib: A Drug Review

open access: yesIndian Journal of Medical and Paediatric Oncology
REarranged during Transfection (RET) is a transforming proto-oncogene that codes for the tyrosine kinase receptor. Pralsetinib is an orally bioavailable, selective inhibitor of mutant forms and fusions involving the RET proto-oncogene.

A Hybrid Bioprinting–Microfabrication Approach to Controllable Multitype Spheroid Arrays for Modeling Tumor Heterogeneity

open access: yesAdvanced Healthcare Materials, EarlyView.
This work introduces a “cancer‐in‐a‐well” technology that recreates tumor heterogeneity in vitro using multitype spheroid arrays. Combining bioprinting/microdispensing with microstructured culture devices, the system improves drug screening and modeling of tumor complexity.

Monitoring pharmacodynamic and molecular drug targets in liquid biopsy: Exploratory study in liver cancer with modelling of EGFR Receptor engagement

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aim Liquid biopsy is minimally invasive (compared with tissue biopsy) and has previously been used to generate systems data regarding drug elimination via hepatic enzymes and transporters. This study extends quantitative assessment of systems parameters in liquid biopsy to pharmacodynamic (PD) and disease markers relevant to cancer development
