Short‐Term Severe Energy Restriction Promotes Molecular Health and Reverses Aging Signatures in Adults With Prediabetes in the PREVIEW Study

Severe short‐term energy restriction in adults with prediabetes led to a significant reduction in circulating proteins linked to inflammation, senescence, and protein turnover. These changes, partly mediated by extracellular vesicles, suggest systemic dietary‐induced benefits beyond glycemic control, including improved aging biomarkers.

Senescence Alters Antimicrobial Peptide Expression and Induces Amyloid‐β Production in Retinal Pigment Epithelial Cells

Healthy RPE cells produce AMPs and AMP precursors, including amyloid precursor protein (APP) and chemerin, and protect the neuroretina from blood‐borne pathogens and maintain homeostasis. When RPE cells undergo senescence, the antibacterial activity declines, and APP undergoes amylogenesis, leading to Aβ deposition.

βA3/A1‐crystallin regulates HDAC3 activity and epigenetic processes in the RPE. (A) The illustration demonstrates that βA3/A1‐crystallin plays a crucial role in maintaining IPMK protein expression, which in turn activates HDAC3. (B) When βA3/A1‐crystallin is absent (Cryba1 KO ), IPMK protein expression decreases, resulting in the deactivation of HDAC3.
