Fine-mapping identifies multiple prostate cancer risk loci at 5p15, one of which associates with TERT expression [PDF]

open access: yes, 2013
Associations between single nucleotide polymorphisms (SNPs) at 5p15 and multiple cancer types have been reported. We have previously shown evidence for a strong association between prostate cancer (PrCa) risk and rs2242652 at 5p15, intronic in the ...

Zinc Exposure Causes Disulfidptosis to Induce Miscarriage by Up‐Regulating GATA1/METTL1/SLC7A11 Axis

open access: yesAdvanced Science, EarlyView.
Zn exposure up‐regulates GATA1, promoting GATA1‐mediated METTL1 and SLC7A11 transcription. It also enhances METTL1‐mediated m7G modification on SLC7A11 mRNA, increasing SLC7A11 mRNA stability. Ultimately, Zn exposure up‐regulates SLC7A11 at both transcriptional and post‐transcriptional levels, causing disulfidptosis. Knockdown of murine Slc7a11, Gata1,

A Radioresistant‐Tumor‐Targeted Nanoparticle for X‐Ray‐Controlled Nitric Oxide Release to Potentiate Radiotherapy

open access: yesAdvanced Science, EarlyView.
The PBTN targets GRP78 and, upon X‐ray irradiation, generates ONOO− to induce immunogenic cell death, thereby activating CD8+ T cells and establishing a systemic antitumor immune response. ABSTRACT Nitric oxide (NO) treated radioresistant tumors by relieving hypoxia and blocking DNA repair, but its nonselective toxicity has precluded therapeutic use ...

Combating VEGFA‐siRNA‐Induced Metabolic Reprogramming via Glucose Utilization Deprivation

open access: yesAdvanced Science, EarlyView.
An ionizable lipid nanoparticle co‐delivers VEGFA siRNA and glucose oxidase to tumors to counteract VEGFA knockdown‐induced, glutamine‐driven metabolic compensation. Enzymatic glucose consumption reverses the adaptive pathways and restores metabolism toward baseline, thereby sensitizing tumors to therapy.

HTLV-1 reverse transcriptase homology model provides structural basis for sensitivity to existing nucleoside/nucleotide reverse transcriptase inhibitors

open access: yesVirology Journal
The human T-lymphotropic virus type 1 (HTLV-1) infects millions of people globally and is endemic to various resource-limited regions. Infections persist for life and are associated with increased susceptibility to opportunistic infections and severe ...

Molecular Glue cc‐885 Inhibits VHL‐Deficient Clear Cell Renal Cell Carcinoma via ETS1 Degradation

open access: yesAdvanced Science, EarlyView.
VHL‐deficient kidney cancer lacks effective treatments. This study reveals that the molecular glue degrader cc‐885 hijacks the cellular recycling system to selectively destroy the oncogenic protein ETS1, effectively killing VHL‐mutant tumors. Combining CC‐885 with the approved drug belzutifan achieves powerful synergy, offering a promising new ...

One-Step Fluorescent Probe Product-Enhanced Reverse Transcriptase Assay

open access: yesBioTechniques, 1998
Recently developed PCR-based reverse transcriptase (RT) assays are useful in the detection of retroviruses since they are approximately a millionfold more sensitive than conventional RT assays.

Methoxyflavones from as HIV-1 Reverse Transcriptase Inhibitors

open access: yesNatural Product Communications, 2017
Methoxyflavones are flavonoid widely distributed in plants and has been reported as potent antitumor agents and some of them have shown activity against HIV-1. In this work, two methoxyflavones isolated from Marcetia taxifolia were evaluated in vitro and

Asymmetric conformational maturation of HIV-1 reverse transcriptase

open access: yeseLife, 2015
HIV-1 reverse transcriptase utilizes a metamorphic polymerase domain that is able to adopt two alternate structures that fulfill catalytic and structural roles, thereby minimizing its coding requirements. This ambiguity introduces folding challenges that
