Peptide Substrates for Rho-Associated Kinase 2 (Rho-Kinase 2/ROCK2)

open access: yesPLoS ONE, 2011
Peptide substrates sensitive for a certain protein kinase could be important for new-drug development and to understand the mechanism of diseases. Rho-associated kinase (Rho-kinase/ROCK) is a serine/threonine kinase, and plays an important part in cardiovascular disease, migration and invasion of tumor cells, and in neurological disorders.

Enhancing Maturation of Human Neuromuscular Organoids via Electrical Stimulation

open access: yesAdvanced Science, EarlyView.
A framework for on‐demand and non‐invasive exposure of human neuromuscular organoids (NMOs) to electrical stimuli is established to promote their maturation. The robustness and effectiveness of different stimulation regimes are evaluated via thorough characterization of organoid tissue structure and contraction capacity. Chronic electrical stimulation,

CALB2 is a Mechanoresistance Gene in Metastatic Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Prostate cancer cell lines of differing origins were cultured over many passages while being subjected to semi‐lethal levels of fluid shear stress, representing the harsh mechanical environment these cells are subjected to in the circulation during the process of distant metastasis.

The Rho-associated kinase inhibitor fasudil can replace Y-27632 for use in human pluripotent stem cell research.

open access: yesPLoS ONE, 2020
Poor survival of human pluripotent stem cells (hPSCs) following freezing, thawing, or passaging hinders the maintenance and differentiation of stem cells. Rho-associated kinases (ROCKs) play a crucial role in hPSC survival.

Human Dental Pulp Stem Cell Secretome Restores Ischemic Stroke–Impaired Motor and Cognitive Functions by Reprogramming Redox and Inflammatory Signaling

open access: yesAdvanced Science, EarlyView.
Human dental pulp stem cell secretome emerges as a cell‐free therapeutic strategy for ischemic stroke by reprogramming redox and inflammatory signaling. hDPSC secretome‐derived antioxidant and immunomodulatory factors suppress the TLR4–NOX–ROS–NF‐κB axis, reduce microglial inflammatory responses and apoptosis, and promote neurogenesis, angiogenesis ...

EDNRA Forms a Positive Feedback Loop with the Hippo/YAP Axis to Drive Triple‐Negative Breast Cancer Progression

open access: yesAdvanced Science, EarlyView.
Endothelin receptor type A (EDNRA) and the Hippo/YAP pathway form a self‐reinforcing loop that sustains triple‐negative breast cancer. EDNRA activates YAP through Gαq/11–Rho/ROCK–LATS signaling, while YAP/TEAD4 reciprocally drives EDNRA transcription.

ROCK activity regulates functional tight junction assembly during blastocyst formation in porcine parthenogenetic embryos [PDF]

open access: yesPeerJ, 2016
The Rho-associated coiled-coil-containing protein serine/threonine kinases 1 and 2 (ROCK1 and ROCK2) are Rho subfamily GTPase downstream effectors that regulate cell migration, intercellular adhesion, cell polarity, and cell proliferation by stimulating ...

Chromatin Packing Domain Engineering Through the Manipulation of Nuclear Cationic States

open access: yesAdvanced Science, EarlyView.
Manipulation of intranuclear divalent cation concentrations rapidly and reversibly remodels chromatin packing domains in living cells. Magnesium depletion disrupts domain compaction, heterochromatin organization, and transcriptional regulation, whereas magnesium loading promotes more compact and stable domains.

Extracellular CIRP Dysregulates Microglial Efferocytosis in Acute Ischemic Stroke via the TLR4/miR‐155/MafB Axis

open access: yesAdvanced Science, EarlyView.
eCIRP released in the stroke brain binds to TLR4 expressed on microglia to induce miR‐155, which suppresses MafB, decreases MerTK and downstream signaling to impair efferocytosis of apoptotic neurons, worsening the acute stroke outcomes. Inhibition of eCIRP‐TLR4 interaction with small peptide C23 attenuates eCIRP‐induced microglial efferocytic ...

Rho-related GTP-binding protein RhoE (RND3) regulates multiple myeloma bone disease

open access: yesCancer Cell International
Multiple Myeloma bone disease (MMBD) is highly prevalent in the multiple myeloma (MM) patients. Using the patients’ gene expression profile, we identified Rho Family GTPase 3 (RND3) as a gene whose expression in MM negatively associated with MMBD ...
