Small nuclear RNAs in messenger RNA and ribosomal RNA processing.
The FASEB Journal, 1993Ribonucleoproteins (RNPs) play essential roles in many aspects of gene expression. Two families of nuclear RNPs are involved in the processing of primary transcripts made by RNA polymerases I and II (pol I and II), two of the three polymerases present in the nuclei of eukaryotic cells.

Formation of Pseudouridine in U5 Small Nuclear RNA
Biochemistry, 1994The formation of pseudouridine (psi) on U5 small nuclear RNA (U5 snRNA) was studied using an in vitro modification system. Labeled U5 RNA, synthesized in vitro and therefore unmodified, was incubated in reactions containing S100 and/or nuclear extracts (NE) from HeLa cells, and the levels of psi were determined.

Small nuclear RNAs and pre-mRNA splicing
Current Opinion in Cell Biology, 1994Genetic and biochemical studies have recently shown that the splice sites in nuclear pre-mRNAs are aligned for the transesterification reactions through a collaboration between U5 and U6 small nuclear RNAs, which form the heart of a network of RNA-based interactions in the active spliceosome.

Transcription of a gene for human U1 small nuclear RNA
Cell, 1982Transcription of U1 small nuclear RNA from a 592 bp fragment of human DNA was analyzed in vivo and in vitro. When injected into Xenopus laevis oocyte nuclei, the cloned DNA is transcribed by RNA polymerase II to make human U1 snRNA. Thus the sequences of this fragment are sufficient for expression of the U1 snRNA gene.

Stimulation of transcription of chromatin by specific small nuclear RNAs
Gene, 1980Small molecular weight nuclear RNAs (SnRNA) purified from the chromatin of SV40-transformed W138 human fibroblasts have been found to stimulate transcription of chromatin in homologous isolated nuclei as well as in nuclei of untransformed human and monkey cells. Stimulation in normal cell nuclei involves an increase in both initiation sites and rate of

Specific small nuclear RNAs are associated with yeast spliceosomes
Cell, 1986Two different methods have been devised for the analysis and purification of spliceosomes formed in a yeast in vitro splicing system. The first method relies on the electrophoretic separation of ribonucleoprotein particles in composite acrylamide-agarose gels. A large fraction of added substrate is located in spliceosomes, the formation of which can be

Nuclear RNA splicing and small nuclear RNAs and their genes in higher plants
1991Abstract This review is focused on the splicing of nuclear precursors of messenger and transfer RNAs, and on the synthesis and structure of the U small nuclear RNAs (U- snRNAs), which participate in various RNA processing events in the nucleus.

Small nuclear rnas: An association with condensed chromatin in amoebae
Journal of Cell Science, 1978ABSTRACTFor the purpose of studying the relationship of small nuclear RNAs (snRNAs) to chromatin, we studied the effect of the chromatin-binding agent actinomycin D (AMD) on the nuclei of amoebae. By electron microscopy we noted that when mitotic cells were placed in AMD their chromatin was highly condensed 3 h later. This contrasts strikingly with the

Publisher Summary The snRNAs exist in the cell as ribonucleoprotein (RNP) complexes. While the snRNAs U1, U2, and U5 are organized in separate RNP particles, the majority of the snRNAs U4 and U6 reside in a single RNP complex. Several proteins in the molecular weight range of 25K to 200K were recently identified as possible U5-specific proteins ...

Small Nuclear RNAs: RNA Sequences, Structure, and Modifications
1988All eukaryotic cells contain multiple small nuclear RNAs, designated U-sn At present ten U-snRNAs have been identified; they account for about 1% total mammalian cellular RNA. Of these, the U1 to U8 snRNAs have been sively characterized. All the U-snRNAs contain a 5′-cap structure consis a blocked 5′-terminal pyrophosphate linkage; except for U6 RNA, a
