ROSIGLITAZONE AND CARDIOVASCULAR RISK – A REVIEW
The thiazolidinediones (TZDs) are a class of oral drugs used for the management of type 2 diabetes mellitus and act as ligands for the transcription factor Peroxisome Proliferator-Activated Receptor gamma (PPARγ).

ABSTRACT Post‐translational modifications (PTMs) play a critical role in regulating the transcriptional activity of PPARγ, a nuclear receptor central to glucose and lipid homeostasis. Among these, lysine acetylation at K268 and K293 and phosphorylation at S273 are particularly relevant to insulin sensitivity.

Rosiglitazone: to be or not to be? [PDF]
To the Editor: The American Diabetes Association (ADA) and the EASD recently recommended an update [1] to their algorithm for the initiation and adjustment of therapy for the management of hyperglycaemia in type 2 diabetes, which was originally published in 2006 [2] and specifically revised in early 2008 regarding the use of thiazolidinediones [3]. Sexual Safety Signals of Common Antidiabetic Drugs: Insights From FAERS Disproportionality Analysis
ABSTRACT Background Erectile dysfunction is a prevalent and distressing complication of diabetes mellitus that markedly affects quality of life in male patients. Although the role of diabetes in erectile dysfunction is well‐established, potential drug‐induced sexual dysfunction associated with commonly prescribed antidiabetic medications remains ...

Pioglitazone is a valid alternative to rosiglitazone
In 2000, two new thiazolidinediones, pioglitazone and rosiglitazone, were approved for the treatment of type 2 diabetes mellitus (T2DM). Administered as monotherapy or in combination with other antihyperglycemic agents, they represented an effective ...

PDE4D and PDE3B orchestrate distinct cAMP microdomains in 3T3‐L1 adipocytes
Basal conditions: •Ins/PDE3B lowers cytoplasmic cAMP (cyt‐cAMP) without affecting plasma membrane cAMP (pm‐cAMP). •Insulin decreases lipid droplet cAMP (LD‐cAMP) independent of PDE3B. •FGF1/PDE4D modestly reduces both cyt‐ and pm‐cAMP, while PDE4D alone can modulate LD‐cAMP. ISO stimulation: •Ins/PDE3B has minimal impact on cyt‐cAMP.
