Adipocyte‐Derived Palmitic Acid Promotes Breast Cancer Malignancy via ZDHHC15‐Mediated S‐Palmitoylation of PPARγ

open access: yesCancer Science, EarlyView.
Adipocyte‐derived palmitic acid induces ZDHHC15‐mediated PPARγ S‐palmitoylation at Cys159, triggering its proteasomal degradation and promoting breast cancer malignancy with rosiglitazone resistance. Inhibition of PPARγ S‐palmitoylation by 2‐bromopalmitate suppresses breast cancer progression and synergistically enhances rosiglitazone efficacy ...

Effect of rosiglitazone on progression of atherosclerosis: insights using 3D carotid cardiovascular magnetic resonance

open access: yesJournal of Cardiovascular Magnetic Resonance, 2009
Background There is recent evidence suggesting that rosiglitazone increases death from cardiovascular causes. We investigated the direct effect of this drug on atheroma using 3D carotid cardiovascular magnetic resonance.

SGLT‐2 Inhibitors are Associated With a Lower Risk of Multiple Myeloma

open access: yesEuropean Journal of Haematology, EarlyView.
ABSTRACT Aims We aimed to investigate drug response of sodium‐glucose transport protein‐2 inhibitors (SGLT2i) in multiple myeloma (MM), as SGLT2i have potential favorable effects on conditions related to MM. Methods We conducted a pharmacoepidemiologic study with a US nationwide health insurance claim data using Cox models to investigate the ...

Insulin Sensitization by a Novel Partial Peroxisome Proliferator-Activated Receptor γ Agonist With Protein Tyrosine Phosphatase 1B Inhibitory Activity in Experimental Osteoporotic Rats

open access: yesJournal of Pharmacological Sciences, 2014
The pharmacological profile of (S)-7-(2-{2-[(E)-2-cyclopentylvinyl]-5-methyloxazol-4-yl}-ethoxy)-2-[(2E,4E)-hexadienoyl]-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid (KY-201), a peroxisome proliferator-activated receptor (PPAR) γ agonist, was ...

Adipocyte‐specific NIK depletion enhances energy metabolism and glucose tolerance in mice

open access: yesThe FEBS Journal, EarlyView.
Obesity is associated with reduced energy expenditure and increased hepatic lipid accumulation. This study reveals that loss of NIK in adipocytes stimulates FGF21‐driven expression of thermogenic genes, including UCP1, and mitochondrial uncoupled respiration.

Tankyrase‐2 regulates adipocyte differentiation through AMPK/mTOR signaling

open access: yesThe FEBS Journal, EarlyView.
Tankyrase 2 (TNKS2), a PARP family member, underpins adipocyte differentiation. TNKS2 controls LKB1 protein level, and consequently, the activity of the mTOR/AMPK system and adipocyte differentiation through a layered process. Pharmacological inhibition of TNKS2 leaves other PARPs and DNA repair active and therefore represents a novel target in ...

The Effect of PPAR Agonists on the Migration of Mature and Immature Eosinophils

open access: yesPPAR Research, 2012
PPARγ agonists can either enhance or inhibit eosinophil migration, which is a sum of directional migration (chemotaxis) and random cell movement (chemokinesis). To date, the effects of PPAR agonists on chemokinesis have not been examined.

Cardiovascular preventive role of thiazolidinedione in patients with type 2 diabetes mellitus undergoing percutaneous coronary intervention

open access: yesJournal of Diabetes Investigation, EarlyView.
Among patients with type 2 diabetes undergoing percutaneous coronary intervention, thiazolidinedione use was independently associated with a significantly reduced risk of a composite of myocardial infarction, stroke, and all‐cause death, notably without an increased risk of admission for heart failure. ABSTRACT Background Thiazolidinediones (TZD) are a
