Pathway Evolution Through a Bottlenecking-Debottlenecking Strategy and Machine Learning-Aided Flux Balancing. [PDF]

open access: yesAdv Sci (Weinh)
A biofoundry‐assisted strategy for pathway bottlenecking and debottlenecking enables the parallel evolution of all pathway enzymes along a predictable evolutionary trajectory. A machine learning model can further relax the epistasis of the evolved pathway by optimizing the corresponding promoter combinations.

The serine transporter SdaC prevents cell lysis upon glucose depletion in Escherichia coli. [PDF]

open access: yesMicrobiologyopen, 2020
Our results reveal that the serine transporter SdaC plays a critical role in maintaining amino acid homeostasis during shifts in nutrient availability in Escherichia coli. Abstract The amino acid serine plays diverse metabolic roles, yet bacteria actively degrade exogenously provided serine via deamination to pyruvate.

Engineering a Robust Escherichia coli W Platform for Scalable Production of Flavonoid-O-Glucosides. [PDF]

open access: yesMicrob Biotechnol
Escherichia coli W demonstrates enhanced flavonoid tolerance while the optimisation of sucrose metabolism in this strain enables efficient chemical glycosylation. Elevated UDP‐glucose production requires both pathway overexpression and removal of native glucose catabolism.

CRISPR/Cas9 Ablation of Integrated HIV-1 Accumulates Proviral DNA Circles with Reformed Long Terminal Repeats. [PDF]

open access: yesJ Virol, 2021
Gene editing may be used to excise the human immunodeficiency virus type 1 (HIV-1) provirus from the host cell genome, possibly eradicating the infection. Here, using cells acutely or latently infected by HIV-1 and treated with long terminal repeat (LTR)-

Ultrasensitive and Wide‐Range Flexible Hydrogen Sensor Based on Pd Nanoparticles Decorated Ultrathin SnO2 Film

open access: yesAdvanced Electronic Materials, Volume 9, Issue 3, March 2023., 2023
Pd NPs‐decorated ultrathin SnO2 film for hydrogen gas detection at low temperature is prepared on PI. This kind of flexible film hydrogen sensor exhibits superb sensitivity and wide detection range. The combination of atomic layer deposition technology (ALD) and cluster beam deposition technology (CBD) provides a feasible strategy for manufacturing ...

Engineered acetoacetate‐inducible whole‐cell biosensors based on the AtoSC two‐component system

open access: yesBiotechnology and Bioengineering, Volume 118, Issue 11, Page 4278-4289, November 2021., 2021
Rutter and coworkers engineer a range of acetoacetate sensitive whole‐cell biosensors, based on the AtoSC two‐component system. A modelling and sensitivity analysis approach is applied to identify factors that are important in determining the response of these biosensors.

Measuring Colloidal Forces With Atomic Force Microscopy 1: Salt Influence on Hydrophobic and Hydrophilic Interactions. [PDF]

open access: yesMicrosc Res Tech
Colloidal forces play an important role in many life phenomena. Here, we present the preparation of hydrophobic and hydrophilic systems for the study of colloidal forces by AFM. The data are modeled using an extended DLVO model. ABSTRACT Colloidal forces are essential for maintaining the stability and functionality of colloidal systems, affecting ...

SWI/SNF-like chromatin remodeling factor Fun30 supports point centromere function in S. cerevisiae [PDF]

open access: yes, 2011
Budding yeast centromeres are sequence-defined point centromeres and are, unlike in many other organisms, not embedded in heterochromatin. Here we show that Fun30, a poorly understood SWI/SNF-like chromatin remodeling factor conserved in humans ...

Regulatory activity revealed by dynamic correlations in gene expression noise [PDF]

open access: yes, 2008
Gene regulatory interactions are context dependent, active in some cellular states but not in others. Stochastic fluctuations, or 'noise', in gene expression propagate through active, but not inactive, regulatory links^(1,2).

Effects of the post-spinal cord injury microenvironment on the differentiation capacity of human neural stem cells derived from induced pluripotent stem cells [PDF]

open access: yes, 2016
This work was supported by TERCEL and CIBERNED funds from the Instituto de Salud Carlos III of Spain, and FEDER funds from the EC.Spinal cord injury (SCI) causes loss of neural functions below the level of the lesion due to interruption of spinal ...
