scTIDE: Deciphering Critical Transitions Through Cell‐Perturbed Manifold Graphs and Optimal Transport Conditional Flow Matching

open access: yesAdvanced Science, EarlyView.
scTIDE identifies single‐cell tipping points by combining manifold‐based graph representations with optimal‐transport conditional flow matching, which preserves intrinsic topology and models distributional dynamics. It supports critical‐transition detection at individual‐cell resolution, prediction of unseen cells, and dimensionality reduction and ...

Tyrosine phosphatase SHP2 promoted the progression of CRC via modulating the PI3K/BRD4/TFEB signaling induced ferroptosis

open access: yesDiscover Oncology
Objective To elucidate the mechanism by which tyrosine phosphatase SHP2 protects CRC through modulation of TFEB-mediated ferritinophagy, thereby suppressing ROS and ferroptosis. Methods SW480 and SW620 cells, in the logarithmic growth phase, were treated

Biomaterials for modulating cellular responses of endogenous tendon stem/progenitor cells: A promising avenue for tendon regeneration

open access: yesBMEMat, EarlyView.
This review comprehensively summarizes emerging biomaterial‐based strategies and underlying mechanisms for modulating endogenous tendon stem/progenitor cells (TSPCs). It offers the most recent insights into TSPC physiology and potential applications of tissue engineering and regenerative medicine in tendons.

SHP2 inhibition enhances the anticancer effect of Osimertinib in EGFR T790M mutant lung adenocarcinoma by blocking CXCL8 loop mediated stemness

open access: yesCancer Cell International, 2021
Background Additional epidermal growth factor receptor (EGFR) mutations confer the drug resistance to generations of EGFR targeted tyrosine kinase inhibitor (EGFR-TKI), posing a major challenge to developing effective treatment of lung adenocarcinoma ...

Enhanced peripheral blood mononuclear cell lipid metabolism drives poor immunotherapy response via ferroptotic stress‐induced T cell exhaustion in hepatocellular carcinoma

open access: yesInterdisciplinary Medicine, EarlyView.
Integrated multi‐omics reveals that CD36‐mediated lipid overload in PBMCs drives T cell exhaustion via sub‐lethal ferroptotic stress and AKT suppression. This systemic metabolic signature serves as a robust predictor of immunotherapy response in HCC, suggesting that modulating lipid metabolism can rescue immune fitness.

Single‐Cell Profiling Identifies CLEC5A+ Macrophages as Key Drivers of Thoracic Aortic Aneurysm Via CCL5‐Mediated M1 Polarization

open access: yesJournal of Clinical Laboratory Analysis, EarlyView.
Thoracic aortic aneurysm (TAA) is a life‐threatening cardiovascular disease with limited therapeutic options. Through single‐cell RNA sequencing of aortic tissues from healthy individuals and TAA patients (105,541 cells), we identified CLEC5A+macrophages as the predominant pathogenic cell population exhibiting the highest M1 polarization score. Machine

A Comprehensive Study of Bidirectional Interactions Between the Human Microbiome and Blood Malignancies and Hematologic Conditions: Focus on Novel Therapeutic Strategies

open access: yesJournal of Clinical Laboratory Analysis, EarlyView.
Microbiota‐targeted therapeutic strategies in hematological disorders. aHSCT: Transplanted immune cells from the donor may attack the recipient's tissues, especially when damage to the intestinal epithelium disrupts the gut microbiota, contributing to GVHD. Maintaining a balanced gut microbiota that supports immune regulation helps reduce GVHD risk and

Effect of SHP2 knockdown on the proliferation and osteogenic differentiation of human periodontal ligament stem cells under inflammatory environment

open access: yes口腔疾病防治, 2022
Objective The purpose of this study was to clarify the regulatory effect and mechanism of Src homology-2 domain containing protein tyrosine phosphatase-2 (SHP2) on human periodontal ligament stem cell (hPDLSC) proliferation and osteogenic differentiation

Atomistic ensemble of active SHP2 phosphatase

open access: yesCommunications Biology, 2023
AbstractSHP2 phosphatase plays an important role in regulating several intracellular signaling pathways. Pathogenic mutations of SHP2 cause developmental disorders and are linked to hematological malignancies and cancer. SHP2 comprises two tandemly-arranged SH2 domains, a catalytic PTP domain, and a disordered C-terminal tail.

Muscle‐Specific Kinase Signaling and Its Therapeutic Potential

open access: yesMuscle &Nerve, EarlyView.
ABSTRACT The function of the neuromuscular junction (NMJ) is compromised in many neuromuscular diseases (NMDs) such as autoimmune or congenital myasthenia gravis (MG), amyotrophic lateral sclerosis (ALS), spinal muscular atrophy (SMA), and muscular dystrophies.
