Robust Single-Particle Cryo-Em Image Denoising and Restoration

open access: yesICASSP 2024 - 2024 IEEE International Conference on Acoustics, Speech and Signal Processing (ICASSP)
Cryo-electron microscopy (cryo-EM) has achieved near-atomic level resolution of biomolecules by reconstructing 2D micrographs. However, the resolution and accuracy of the reconstructed particles are significantly reduced due to the extremely low signal-to-noise ratio (SNR) and complex noise structure of cryo-EM images.

Enhancing electrospray ionization efficiency for particle transmission through an aerodynamic lens stack

open access: yesJournal of Synchrotron Radiation
This work investigates the performance of the electrospray aerosol generator at the European X-ray Free Electron Laser (EuXFEL). This generator is, together with an aerodynamic lens stack that transports the particles into the X-ray interaction vacuum ...

Single-shot femtosecond x-ray diffraction from randomly oriented ellipsoidal nanoparticles

open access: yesPhysical Review Special Topics. Accelerators and Beams, 2010
Coherent diffractive imaging of single particles using the single-shot “diffract and destroy” approach with an x-ray free electron laser (FEL) was recently demonstrated.

Hydrostatic pressure activates HIF‐1α via β‐catenin to promote stemness in breast cancer cells

open access: yesFEBS Open Bio, EarlyView.
To mimic the elevated intestinal fluid pressure in breast cancers, we loaded human breast cancer cells (MCF‐7, MDA‐MB‐453, and BT‐474) to 50 mmHg hydrostatic pressure. Hydrostatic pressure exposure upregulated HIF‐1α and induced stemness in MCF‐7 and BT‐474 cells.

Imaging Live Cells at the Nanometer-Scale with Single-Molecule Microscopy: Obstacles and Achievements in Experiment Optimization for Microbiology

open access: yesMolecules, 2014
Single-molecule fluorescence microscopy enables biological investigations inside living cells to achieve millisecond- and nanometer-scale resolution. Although single-molecule-based methods are becoming increasingly accessible to non-experts, optimizing ...

The crystal structure of the Borrelia burgdorferi nicotinamidase BBE22 resolves a long‐standing annotation error

open access: yesFEBS Open Bio, EarlyView.
The crystal structure of Borrelia burgdorferi nicotinamidase (PncA/BBE22) reveals the correct full‐length protein initiated from a non‐canonical AUU start codon. The structure validates previous biochemical findings and resolves a long‐standing annotation error, demonstrating that the truncated database sequence is structurally incompatible with the ...

A minimal view of single-particle imaging with X-ray lasers [PDF]

open access: yesPhilosophical Transactions of the Royal Society B: Biological Sciences, 2014
The ability to serially interrogate single biomolecules with femtosecond X-ray pulses from free-electron lasers has ushered in the possibility of determining the three-dimensional structure of biomolecules without crystallization. However, the complexity of imaging a sample's structure from very many of its noisy and incomplete diffraction data can be ...

Evaluating the involvement of autolysosomes in the nuclear translocation of fluorescent proteins

open access: yesFEBS Open Bio, EarlyView.
Endogenously expressed fluorescent proteins can be degraded by autophagy and transported to cell nuclei via the nuclear pore complex. But in some cell lines, for example, HeLa cells which are positive for immunoreactivity of a receptor ligand, such as UCN I, in cell nuclei, fusion of autolysosome with the nuclear envelope is involved in the nuclear ...

Introducing a frequency-tunable magnetic particle spectrometer

open access: yesCurrent Directions in Biomedical Engineering, 2015
Image quality in the new imaging modality magnetic particle imaging (MPI) heavily relies on the quality of the magnetic nanoparticles in use. Therefore, it is crucial to understand the behaviour of such particles.
