Sodium Saccharin Inhibits Adenylyl Cyclase Activity in Non-Taste Cells

We have studied the in vitro effect of sodium saccharin (NaSacch) on the rat adipocyte adenylyl cyclase complex. NaSacch (2.5-50 mM) inhibited significantly in a dose-dependent manner basal and isoproterenol-stimulated cAMP accumulation on isolated rat adipocytes.

The in vitro effects of saccharin were evaluated in a transformed rat-bladder epithelial cell line. AY-27 cells were seeded in a 35-mm tissue culture well and incubated for 24 hr with saccharin or other test compounds dissolved in the media. The cytotoxic effects of saccharin were dependent on the salt form tested.

A reversed-phase high-performance liquid chromatographic procedure for the determination of o-toluenesulfonamide and p-toluenesulfonamide in saccharin and saccharin sodium is described.
