Immunoreactive Somatomedin A in Human Serum*

A RIA has been developed for somatomedin A (SM-A) utilizing Sepharose-bound antibodies. This assay, measuring SM-A, the insulin-like growth factors 1 and 2, and somatomedin C, allows determination in serum samples. In comparison with a serum standard, the mean serum levels in patients with acromegaly or GH deficiency and healthy subjects were 8.7 %/- 0.

Diabetics may have normal somatomedins by radioimmunoassay yet decreased somatomedin activity by bioassay. The discrepancy appears due to circulating inhibitory factors; inhibitors in whole diabetic serum antagonize the action of both somatomedins and insulin via noncompetitive interactions.

A RAPID SIMPLIFIED BIOASSAY FOR SOMATOMEDIN

A simplified, more rapid bioassay for the estimation of somatomedin activity, based on the porcine costal cartilage bioassay, is described. A micro-scale system based on the porcine costal cartilage assay is also described. These assay systems showed no decrease in sensitivity or precision, compared with other methods, as a result of the modifications.

Although somatomedin levels may be low in animals with experimental diabetes, values in humans have generally been normal. Because humans with severely decompensated diabetes have not been studied, we characterized somatomedin C responses during metabolic restoration in 21 patients with diabetic ketoacidosis.
