FAK Mediated Coordination of Extracellular Matrix Remodeling and Cellular Mechanics Impairs Nanomedicine Delivery in Solid Tumors

open access: yesAdvanced Science, EarlyView.
FAK orchestrates a mechanical barrier limiting nanomedicine accumulation in solid tumors by coupling ECM remodeling with cytoskeletal tension. Genetic or pharmacological FAK inhibition dismantles this barrier, enhances clathrin‐mediated uptake and penetration, and overcomes delivery limitations to improve antitumor efficacy.

Mechanism‐Informed Language Modeling and Oxygenated 3D Screening Identify Berberine–Enzalutamide Synergy in Prostate Cancer Models

open access: yesAdvanced Science, EarlyView.
By integrating knowledge‐augmented artificial intelligence with an oxygen‐supplemented 3D screening platform, a synergistic interaction between berberine and enzalutamide is identified. The framework integrates compound mechanisms of action with disease‐relevant signaling context to prioritize candidate combinations and generate testable pathway‐based ...

Ultrasound‐Activated Ce(IV)‐Integrated Biomimetic Nanoplatform for Oxygenation‐Reinforced Gas‐Sonodynamic Ferroptosis and cGAS–STING‐Driven Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
A Ce(IV)‐integrated biomimetic nanoplatform coordinates tumor oxygenation, ultrasound‐triggered carbon monoxide release, sonodynamic therapy, ferroptosis amplification, and cyclic GMP–AMP synthase (cGAS)–stimulator of interferon genes (STING) immune activation.

Low‐Density Lipoprotein Receptor‐Targeted Biomimetic Nanoplatform Suppresses Glioblastoma Stemness and Remodels Antitumor Immunity via Dual Metabolic Disruption

open access: yesAdvanced Science, EarlyView.
A low‐density lipoprotein receptor‐oriented biomimetic nanoplatform couples brain‐tumor delivery with a mechanistically matched regorafenib‐erythrodiol combination. Concurrent disruption of autophagic nutrient salvage and cholesterol homeostasis collapses bioenergetic and signaling support, suppresses glioblastoma stemness, and promotes immunogenic ...
