A Living Multi‐Omics PDX Biobank of High‐Risk Sarcomas Reveals ecDNA‐Driven Oncogene Amplification in Osteosarcoma and Enables Preclinical Precision Oncology

open access: yesAdvanced Science, EarlyView.
A living multi‐omics sarcoma PDX biobank is enriched for high‐risk disease and preserves key patient tumor features at histopathological and molecular levels. Integrated profiling reveals ecDNA‐driven oncogene amplification in osteosarcoma and preferential retention of ecDNA during PDX evolution.

A Biomimetic cRGDyk‐Engineered Platelet Nanoplatform for ACAD11 Delivery to Trigger Pyroptosis in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
A biomimetic platelet nanoplatform (OE‐ACAD11@R‐PLT) is developed for targeted hepatocellular carcinoma immunotherapy. By selectively delivering therapeutic plasmids, this system induces mitochondrial dysfunction and triggers robust NLRP3/Caspase‐1/GSDMD‐mediated pyroptosis in tumor cells.

Near‐Infrared Triggered Rechargeable Singlet Oxygen Battery for Sustained Phototherapy: Fast Charging and Long‐Lasting Discharging

open access: yesAdvanced Science, EarlyView.
A near‐infrared‐triggered rechargeable singlet oxygen battery based on PBDP nanoparticles is developed. Upon 808 nm irradiation for 5 min, the system enables efficient singlet oxygen generation, reversible storage via cycloaddition, and sustained release for up to 1 h after light cessation.

Fc‐Dependent Targeting of the GAS6–MERTK Axis With Monoclonal and Bispecific Antibodies Overcomes Drug Resistance Across EGFR Mutations in Exons 19, 20, and 21

open access: yesAdvanced Science, EarlyView.
EGFR mutations drive a substantial fraction of non‐small cell lung cancers (NSCLC) and are effectively targeted by tyrosine kinase inhibitors (TKIs), including osimertinib. However, resistance inevitably develops through secondary mutations and compensatory receptor activation, including the GAS6 receptor MERTK.
