Nature Immunology, 2019Activation of STING requires its translocation from the endoplasmic reticulum (ER) through the Golgi to vesicles, but the mechanisms that control its localization have been unclear. The ER calcium sensor STIM1 acts as an ER-retention factor that anchors STING on the ER and limits STING signaling.

STIM1, a Calcium sensor localizes mostly to the ER membrane in interphase under resting conditions. Following Ca2+ store depletion, STIM1 forms puncta that localize to the cortical ER and binds Orai1, a Ca2+ channel to allow Ca2+ influx.
