Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

An unexpected insight into the cause of olfactory dysfunction: fibrillogenesis of odorant-binding proteins

open access: yesCell Death Discovery
Olfactory dysfunction is a common complication of serious pathologies, including neurodegenerative disorders, bacterial and viral infections, including COVID-19, and others. Despite the widespread prevalence of olfactory disorders, the pathophysiological

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Elasto-plastic structural stability of arches

open access: yes, 2000
The elastic-plastic structural stability behaviour of arches is analysed in the present work. The application of the developed mathematical model, allows to determine the elastic-plastic equilibrium paths, looking for critical points, bifurcation or limit,

Structural basis of Fanconi anemia pathway activation by FANCM

open access: yesThe EMBO Journal
FANCM is crucial in genome maintenance, functioning in the Fanconi anemia (FA) pathway, alternative lengthening of telomeres (ALT), and replication fork protection.

Structurally unstable quadratic vector fields of codimension two: families possessing a finite saddle-node and an infinite saddle-node

open access: yesElectronic Journal of Qualitative Theory of Differential Equations, 2021
In 1998, Artés, Kooij and Llibre proved that there exist 44 structurally stable topologically distinct phase portraits modulo limit cycles, and in 2018 Artés, Llibre and Rezende showed the existence of at least 204 (at most 211) structurally unstable ...

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

RPAP3 provides a flexible scaffold for coupling HSP90 to the human R2TP co-chaperone complex

open access: yesNature Communications, 2018
The R2TP/PFDL co-chaperone facilitates assembly of RNA polymerase II and PI3-kinase-like kinases such as mTOR by a so far unknown mechanism. Here authors provide the cryo-EM structure of human R2TP, which shows how RPAP3 serves as a flexible platform to ...

Valosin‐containing protein counteracts ATP‐driven dissolution of FUS condensates through its ATPase activity in vitro

open access: yesFEBS Letters, EarlyView.
Biomolecular condensates formed by fused in sarcoma (FUS) are dissolved by high ATP concentrations yet persist in cells. Using a reconstituted system, we demonstrate that valosin‐containing protein (VCP), an AAA+ ATPase, counteracts ATP‐driven dissolution of FUS condensates through its D2 ATPase activity.

Experimental Study on the Thermal Stability of Aluminum Alloy 7075-T651 Structural Parts after Rolling Correction

open access: yes, 2023
The rolling correction process can eliminate machining distortions of aluminum alloy 7075-T651 structural parts. The thermal stability of the corrected structural parts under the action of temperature loading, especially the macroscopic shape stability ...
