Non-covalent interaction between nucleocapsid protein of Tula hantavirus and small ubiquitin-related modifier-1, SUMO-1

To find cellular binding counterparts for the nucleocapsid protein (N) of Tula hantavirus (TULV), two cDNA libraries were screened using yeast two-hybrid systems based on LexA and Gal4 transcription factors. Five cDNA clones encoding SUMO-1 (Small Ubiquitin-related MOdifier, also known as sentrin) were selected in the LexA system.

The small ubiquitin-related modifier-1 (SUMO-1) with broad cellular expression has been implicated in a range of cellular processes, such as cell proliferation, differentiation, and apoptosis. As shown recently, SUMO-1 is expressed and regulated by gonadotropins, in particular an ovulatory hCG stimulus in mouse granulosa cells in vivo.

Examining the Potential Modification of the Protein Tyrosine Kinase Pyk2 by SUMO-1

Proline-rich tyrosine kinase 2 (Pyk2) is a non-receptor protein tyrosine kinase that is highly expressed in hematopoietic cells. Our lab generated two polyclonal antibodies to investigate the regulation of Pyk2 in macrophages and T cells.
