Prescriptions and Ingredients for Controlled CC Bond Formation

open access: yes
Merits and drawbacks of known C-C linking procedures, such as condensation, addn., and isomerization reactions, are outlined. Two novel methods are discussed in more detail: the Cu-catalyzed alkylation of Grignard reagents, and reactions with ...

Does Differentially Private Synthetic Data Lead to Synthetic Discoveries? [PDF]

open access: yes
Background Synthetic data have been proposed as a solution for sharing anonymized versions of sensitive biomedical datasets. Ideally, synthetic data should preserve the structure and statistical properties of the original data, while protecting the ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

The synthetic control method compared to difference in differences: discussion [PDF]

open access: yes, 2014
This presentation was a discussion in response to “The synthetic control method compared to difference-in-differences for evaluating pay-for-performance schemes in health care” presented by Noemi Kreif (LSHTM).

USP29‐regulated noncanonical stabilization of the hypoxia‐inducible factor‐α in aggressive prostate cancer

open access: yesMolecular Oncology, EarlyView.
We identify USP29 as the only DUB mirroring CA9 expression, a marker of hypoxia and HIF pathway activation associated with PCA aggressiveness. USP29 stabilizes HIF‐1α and HIF‐2α via a noncanonical mechanism that is independent of PHD/pVHL activity yet relies on proteasomal regulation, establishing USP29 as a previously unrecognized regulator of hypoxic
