CCL3 and IL‐7 Synergistically Enhance CAR‐T Efficacy in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Synergistic delivery of CCL3 and IL‐7 by CAR‐T cells overcomes the immunosuppressive tumor microenvironment by enhancing cell infiltration, survival, and local immune reprogramming. This also establishes positive feedback that amplifies anti‐tumor activity and memory responses in both CAR‐T and endogenous T cells, presenting a robust therapeutic ...

Development of a CD8 co-receptor independent T-cell receptor specific for tumor-associated antigen MAGE-A4 for next generation T-cell-based immunotherapy

open access: yesJournal for ImmunoTherapy of Cancer, 2021
Background The cancer-testis antigen MAGE-A4 is an attractive target for T-cell-based immunotherapy, especially for indications with unmet clinical need like non-small cell lung or triple-negative breast cancer. Methods An unbiased CD137-based sorting ...

Tumor‐Specific Delivery of CD28 siRNA via Lyso‐PC C‐16 Modified Lipid Nanoparticles Overcomes Anti‐PD‐1 Resistance by Remodeling Tumor Microenvironment

open access: yesAdvanced Science, EarlyView.
This study develops 16:0 LPC‐modified lipid nanoparticles (LPC‐LNPs) with cancer cell specificity by exploiting altered tumor lipid metabolism. LPC‐LNPs encapsulating Cd28 small interfering RNA (LPC‐LNP‐Cd28) knock down cancer cell CD28 without affecting T cells, inflame the tumor microenvironment, and overcome anti‐PD‐1 resistance.

Geometrically repatterned immunological synapses uncover formation mechanisms [PDF]

open access: yes, 2006
The interaction of T cells and antigen-presenting cells is central to adaptive immunity and involves the formation of immunological synapses in many cases.

Antigen Spreading via Localized Administration Enhances Adoptive TCR‐T Cell Therapy in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
Cancer vaccines face limitations in pancreatic cancer due to insufficient tumor antigens. This ionic liquid ILvax via local tumor delivery triggers robust cDC1‐dependent systemic anti‐tumor immunity. It boosts endogenous CD8+ T cells and optimizes adoptive TCR‐T function, providing a promising synergistic therapeutic strategy.

TCR-T cell therapy for solid tumors: challenges and emerging solutions

open access: yesFrontiers in Pharmacology
With the use of T cell receptor T cells (TCR-T cells) and chimeric antigen receptor T cells (CAR-T cells), T-cell immunotherapy for cancer has advanced significantly in recent years.

Oligoclonal expansions of CD8(+) T cells in chronic HIV infection are antigen specific [PDF]

open access: yes, 1998
Acute HIV infection is associated with a vigorous immune response characterized by the proliferation of selected T cell receptor V beta (BV)-expressing CD8(+) T cells.

Inhibition of KDEL Receptors Remodels the Tumor Microenvironment for T Cell Independent Tumor Regression

open access: yesAdvanced Science, EarlyView.
Inhibition of KDELR2 in a small fraction of tumor cells generates sustainable immunogenic cell death conditions within the tumor microenvironment. These conditions promote the regression of tumors in a T cell independent manner. During regression, macrophages prime T cells that subsequently provide systemic protection against recurrence. The potency of

Genetic engineering of T-cell receptors: TCR takes to titin

open access: yesBlood, 2013
In this issue of Blood, Linette et al report a cautionary tale—a lethal off-target toxicity following T-cell receptor (TCR) optimization to enhance affinity against a tumor-associated antigen, MAGE-A3.

Investigating the roles of the alternative Isoforms of the preTCR alpha (pTα) chain in T cell development [PDF]

open access: yes, 2010
PhDThe pre-T cell receptor (preTCR) is required for αβ T cell development and hence efficient adaptive immunity. Composed of a rearranged TCRβ chain paired with the invariant preTCR α chain (pTα), it drives immature thymocytes through the “β-selection ...
