rDNA Copy Number Variation and Methylation During Normal and Premature Aging

open access: yesAging Cell, Volume 24, Issue 5, May 2025.
In > 160 healthy individuals the absolute rDNA copy number (CN) in blood ranged from approximately 240 to 900. The number of active copies with a completely unmethylated promoter varied from 40 to 120, independent of absolute CN. Promoter methylation is a compensation mechanism for absolute CN variation. Aging was associated with a loss of unmethylated

Development of a Short Telomere Zebrafish Model for Accelerated Aging Research and Antiaging Drug Screening

open access: yesAging Cell, EarlyView.
Larvae derived from a cross between tert deficient and haploinsufficient fish show a premature aging phenotype, characterized by telomere attrition and early death. The ST2 larval model is a valuable tool for rapid screening of antiaging molecules. Given that organogenesis occurs within the first 48 h, this model also allows for the conduction of aging

Negative Effect of Gst‐35 on the Health Span of Caenorhabditis elegans Through Lysosomal Dysfunction via the Pmk‐1 and Skr Genes

open access: yesAging Cell, EarlyView.
The overexpression of gst‐35, the mammalian orthologous genes GSTA1, GSTA2, and GSTA3, induces lysosomal dysfunction through the genes pmk‐1 and skr in C. elegans. This overexpression not only promotes an in vivo inflammatory response and accumulation of reactive oxygen species but also significantly shortens the nematode lifespan; inhibits its ...

Immunosenescence Profile Is Associated With Increased Susceptibility to Severe COVID‐19

open access: yesAging Cell, EarlyView.
(A) A previous immunosenescence profile in individuals infected with SARS‐CoV2 creates a permissive environment for progression to severe COVID‐19: higher inflammaging, accumulation of exhausted/senescent T cells, an oligoclonal B‐cell repertoire, and accelerated epigenetic age.

hTERT Increases TRF2 to Induce Telomere Compaction and Extend Cell Replicative Lifespan

open access: yesAging Cell, EarlyView.
In contrast to the dogma that a subset of short telomeres initiates senescence, hTERT variants unable to maintain telomere length block senescence by stabilizing TRF2 via inhibiting upstream E3 ubiquitin ligases. This noncanonical hTERT function also compacts telomeres and blocks senescence‐associated DNA damage signaling. Cancer cells expressing hTERT
