Effects of the hydrophobicity of taxoids on their interaction with tubulin
Bioorganic & Medicinal Chemistry, 2000Modifications of the hydrophobic character at the 7 and 10 positions of the taxoids greatly modified the effect of these drugs on the tubulin microtubule system. The presence of an alkyl chain at these positions decreased the activity while their corresponding more polar analogues restored the activity of these molecules.

Abstract A 70-year-old man was discovered to have melanoma of the right fifth toe. Following toe amputation, he was started on paclitaxel (250 mglm2 ) every 3 weeks, as part of a phase II clinical trial. He received 16 courses with clinical and radiographic disappearance of adenopathy.
