Pathway classification of TCA cycle

IEE Proceedings - Systems Biology, 2006
The structural analysis of large metabolic networks exhibits a combinatorial explosion of elementary modes. A new method of classification has been developed [called aggregation around common motif (ACoM)], which groups elementary modes into classes with similar substructures.

Upcycling the TCA cycle—rewiring tumour-associated fibroblasts

Nature Metabolism, 2021
Alterations to the extracellular matrix have long been associated with cancer progression and therapeutic resistance. Schworer et al. describe a mechanism whereby fibroblasts reroute metabolites to fuel the demands of collagen synthesis, leading to cancer progression.

Regulation of the urea cycle and TCA cycle by ammonia

Advances in Enzyme Regulation, 1966
Abstract This paper presents the biochemical and enzymatic fundamentals of ammonia toxicity and automatic regulation of urea formation from ammonia. 1. 1.It is considered that the oxidation of isocitrate to 2-oxoglutarate is more sensitive to ammonia than the other dehydrogenases in the TCA cycle. 2.

TCA cycle off, ATF4 on for metabolic homeostasis

Trends in Biochemical Sciences, 2022
Tricarboxylic acid (TCA) cycle is a major hub for catabolic and anabolic reactions, yet cellular metabolic adaptations following its inhibition are largely unknown. Using multi-tiered omics approaches, Ryan et al. have shown convergent activation of the integrated stress response (ISR) through ATF4-mediated rewiring of cellular amino acid and redox ...

TCA Cycle Defects in Barth Syndrome

The FASEB Journal, 2016
Barth syndrome (BTHS) is a genetic disorder caused by defective remodeling of the phospholipid cardiolipin (CL) due to mutations in the gene coding for tafazzin. The disorder is characterized by dilated cardiomyopathy and mitochondrial dysfunction. CL is a unique membrane lipid that has multiple cellular functions.

Glutamine drives glucose-independent TCA cycle

Journal of Cancer Science & Therapy, 2012
Methods: We used high-field NMR, in parallel with GC-MS, and direct infusion nanospray FT-ICR-MS as well as stable isotoperesolved metabolomics to trace the fate of 13C and 15N from labeled glucose or glutamine in the P493 human B lymphoma under aerobic, hypoxic glucose-deprived conditions.

The Truncated TCA Cycle in HeLa Cell Mitochondria

1992
A series of studies were carried out on HeLa cell mitochondria to determine (1) the likely pathways for carbon flux from glutamate, (2) the significance of malic enzyme activity, (3) the significance of pyruvate dehydrogenase activity, and finally (4) the significance of the truncated TCA cycle in HeLa cells (Coleman and Lavietes, 1981).

Metabolic engineering: Kick-starting TCA cycling

Nature Plants, 2015
The tricarboxcylic acid cycle has been exhaustively studied for decades so it is not unreasonable to expect that it would retain few undiscovered surprises. However, experimental analyses in cyanobacteria show it to be remarkably plastic, dependent on what it is producing and how much.
