An Imaging‐Guided, Patient‐Specific Guiding Aid (RWNGuide) for Safe and Reproducible Inner Ear Drug Delivery

open access: yesAdvanced Materials Technologies, EarlyView.
A patient‐specific, imaging‐guided aid enables precise and reproducible drug delivery to the inner ear. By guiding therapeutic agents directly to the round window niche, this approach reduces variability in drug localization, improves delivery safety, and addresses a critical bottleneck in inner ear therapy, offering a scalable strategy for precision ...

Gut Microbiota Regulates Systemic Inflammatory Response and Compensatory Anti‐Inflammatory Response Syndromes by Targeting PF4+ Macrophages in Acute Pancreatitis

open access: yesAdvanced Science, EarlyView.
ABSTRACT Acute pancreatitis (AP) begins with pancreatic local inflammation, leading to the onset of systemic inflammatory response syndrome (SIRS), followed by compensatory anti‐inflammatory response syndrome (CARS), which causes immune paralysis and higher mortality rate.

T follicular helper and TH2 cells in allergic responses

open access: yesAllergology International, 2017
IL-4 is a cytokine commonly secreted by TH2 and follicular helper T (TFH) cells after antigenic sensitization. TH2 cells have been thought to be the major contributor of B cell help as a source of IL-4 responsible for class switch recombination to ...

RNA Helicase DDX21 Controls CD4+ T Cell Proliferation and Promotes Inflammatory Bowel Disease via Translational Control

open access: yesAdvanced Science, EarlyView.
ABSTRACT Inflammatory bowel disease (IBD) is characterized by dysregulated T cell responses. RNA helicases, including DExD‐box helicase 21 (DDX21), are pivotal in RNA metabolism, but their role in T cell‐mediated pathology during IBD remains unclear. Here, we demonstrate that DDX21 expression in CD4+ T cells correlates with cell cycle and translation ...

m6A‐Mediated Glycolysis by IL‐37 Drives T Cell Metabolic Reprogramming to Regulate Colitis

open access: yesAdvanced Science, EarlyView.
This study identifies an IL‐37/SIGIRR‐METTL14 regulatory axis that suppresses global m6A modification in CD4+ T cells. IL‐37 signaling, mediated through SIGIRR, inhibits IRAK4 and JNK phosphorylation, leading to downregulation of the methyltransferase METTL14.

Atypical pericapillary Ly6G⁺Nur77⁺ macrophages initiate type-2 immune responses to allergens in the mouse lung

open access: yesNature Communications
The mechanisms of airway allergen sensing and type 2 immune response initiation remain poorly understood. Using a mouse house dust mite (HDM)-induced allergic airway model, we identify a population of lung macrophages located close to alveolar ...

Engineering Immunoregenerative Therapy via an Immunomodulatory Binary Pharmacology Hydrogel Depot for Prolonged Allograft Survival

open access: yesAdvanced Science, EarlyView.
An immunomodulatory hydrogel (iGEL) forms spontaneously upon subcutaneous injection, acting as a tissue‐adhesive depot. It releases anti‐rejection and regenerative agents in response to inflammation, suppressing T‐cell activity, promoting vascular repair, and restoring allograft function without systemic immunosuppression.

Single-Cell RNA Sequencing Reveals T Helper Cells Synthesizing Steroids De Novo to Contribute to Immune Homeostasis

open access: yesCell Reports, 2014
T helper 2 (Th2) cells regulate helminth infections, allergic disorders, tumor immunity, and pregnancy by secreting various cytokines. It is likely that there are undiscovered Th2 signaling molecules.

Engineering Approaches to Modify Immunomodulatory Functions of Mesenchymal Stromal Cells (MSCs): Tissue Regeneration and Clinical Application

open access: yesAdvanced Science, EarlyView.
Mesenchymal stromal cells (MSCs) show promise for treating immune‐related disorders through immunomodulation and tissue regeneration. This review gives a brief overview of current clinical approval of MSC therapies. It also discussed how bioengineering, including genetic modification, biomaterial delivery, extracellular vesicles, and iPSC‐derived MSCs,

Combining network modeling and gene expression microarray analysis to explore the dynamics of Th1 and Th2 cell regulation. [PDF]

open access: yesPLoS Computational Biology, 2010
Two T helper (Th) cell subsets, namely Th1 and Th2 cells, play an important role in inflammatory diseases. The two subsets are thought to counter-regulate each other, and alterations in their balance result in different diseases.
