Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Therapeutics Letter 31

open access: yes, 1999
Therapeutics Letter 31 considers the importance of making unpublished clinical trial data available in the context of the approval of celecoxib. Conclusions: In this case, the only information available to help the clinician decide whether to prescribe ...

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Therapeutics Letter 39

open access: yes, 2001
Therapeutics Letter 39 adds new evidence to information previously published about NSAIDs in Therapeutics Letters 4 and Letter 17. It also responds to our commitment in Therapeutics Letter 31 to report on celecoxib when published trials became available.

Therapeutics 1995

open access: yesJournal of the Royal College of Physicians of London, 1995
A joint conference on therapeutics with the Faculty of Pharmaceutical Medicine was held at the Royal College of Physicians on 28 June 1995, organised by Professor M C L'E Orme. A wide variety of topics was covered ranging from management of acute poisoning to the treatment of myocardial infarction, as well as the teaching of therapeutics in the new ...

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

The role of the HIV-1 gut reservoir in driving early cardiovascular events in people living with HIV

open access: yesFrontiers in Drug Discovery
People with HIV (PWH), even when well-controlled on antiretroviral therapy (ART), are at an increased risk for cardiovascular disease (CVD) and CVD events including sudden cardiac death and acute myocardial infarction (MI).
