9.5 COAGULATION CONTROL BY THE RHOA PATHWAY AND THE EXCHANGE FACTOR ARHGEF1
Platelet activation by thrombin is an auto-amplification loop of thrombin generation, a major factor in the formation of atherosclerotic plaques. The small G protein RhoA, under the direct control of the exchange factor Arhgef1, modulates several ...

Background Patients with mechanical heart valves (MHV) require warfarin to prevent thromboembolism. Although dabigatran was as effective as warfarin for stroke prevention in atrial fibrillation when compared with warfarin in patients with MHV, the study ...

Thrombin generation after cariopulmonary bypass
Thrombin ist das Schlüsselenzym der Gerinnung. Prothrombotische Komplikationen wie Schlaganfälle und Herzinfarkte, sind nach Herzoperationen häufig.

Enhanced thrombin generation after cardiopulmonary bypass surgery.
Thrombin generation has a key role in the pathophysiology of hemostasis. Research has focused on the intraoperative course of hemostasis, while little is known about postoperative hemostatic activation.

Multimodal Imaging Reveals Rapid Catecholamine Uptake and Release by Neutrophils
We show that immune cells (neutrophils) synthesize, uptake, and store catecholamine neurotransmitters such as dopamine or adrenaline. They also release them in response to specific stimuli (serotonin), which we directly visualize using fluorescent nanosensors. We further demonstrate that catecholamines affect neutrophil functions (NETosis) and platelet

Background Low molecular weight heparins (LMWH’s) are used to prevent and treat thrombosis. Tests for monitoring LMWH’s include anti-factor Xa (anti-FXa), activated partial thromboplastin time (aPTT) and thrombin generation.

ABSTRACT Platelet transfusions to treat bleeding complications use donor‐derived platelets stored at room‐temperature, that have a shelf‐life of only 5–7 days due to bacterial contamination risks. Cold‐stored and freeze‐dried platelets are being investigated for extending shelf‐life, but these still have the inherent challenge of donor‐dependency.

Anticoagulants and the Propagation Phase of Thrombin Generation
The view that clot time-based assays do not provide a sufficient assessment of an individual's hemostatic competence, especially in the context of anticoagulant therapy, has provoked a search for new metrics, with significant focus directed at techniques that define the propagation phase of thrombin generation.

Thrombin generation assay (CAT) v1
TG in plasma was measured with the calibrated automated thrombogram (CAT) assay as developed by Hemker and co-workers [18-20]. Briefly, 80 μl platelet poor plasma (PPP) was mixed with 20 μl of a mixture containing tissue factor (Dade-Behring) at a ...
