Cancer‐Like Fragmentomic Characteristics of Somatic Variants in Cell‐Free DNA
We report that in non‐cancerous subjects, cell‐free (cfDNA) molecules harboring somatic variants exhibit cancer‐like fragmentomic characteristics associated with clonal hematopoiesis. Primer length for reverse transcription significantly affects eRNA‐based biodiversity recovery and community structure. Shorter primers, particularly N4 and N5, in random priming and medium‐length primers, particularly T20, in oligo(dT) priming demonstrate superior performance, especially in detecting the rare biosphere.
