Human DNA Polymerase κ Bypasses and Extends beyond Thymine Glycols during Translesion Synthesis in Vitro, Preferentially Incorporating Correct Nucleotides [PDF]

open access: yesJournal of Biological Chemistry, 2002
Human polymerase kappa (polkappa), the product of the human POLK (DINB1) gene, is a member of the Y superfamily of DNA polymerases that support replicative bypass of chemically modified DNA bases (Ohmori, H., Friedberg, E. C., Fuchs, R. P., Goodman, M. F., Hanaoka, F., Hinkle, D., Kunkel, T. A., Lawrence, C.

Epigenotoxicity: Decoding the epigenetic imprints of genotoxic agents and their implications for regulatory genetic toxicology

open access: yesEnvironmental and Molecular Mutagenesis, EarlyView.
Abstract Regulatory genetic toxicology focuses on DNA damage and subsequent gene mutations. However, genotoxic agents can also affect epigenetic marks, and incorporation of epigenetic data into the regulatory framework may thus enhance the accuracy of risk assessment.

Role of Two Strictly Conserved Residues in Nucleotide Flipping and N-Glycosylic Bond Cleavage by Human Thymine DNA Glycosylase [PDF]

open access: yesJournal of Biological Chemistry, 2009
Thymine DNA glycosylase (TDG) promotes genomic integrity by excising thymine from mutagenic G.T mismatches arising by deamination of 5-methylcytosine, and follow-on base excision repair enzymes restore a G.C pair. TDG cleaves the N-glycosylic bond of dT and some other nucleotides, including 5-substituted 2'-deoxyuridine analogs, once they have been ...

Advances in Double‐Stranded DNA Targeting Technologies

open access: yesExploration, EarlyView.
Double‐stranded DNA (dsDNA) plays a crucial role in genetic information storage and disease management, but its inherent stability limits access to internal bases. To overcome this challenge, various high‐specificity molecular targeting technologies have been developed to destabilize the DNA structure.

The Characteristics of Cytochrome C Oxidase Gene Subunit I in Wild Silkmoth Cricula trifenestrata Helfer and Its Evaluation for Species Marker

open access: yesMedia Peternakan, 2012
The study was conducted to assess the characteristics of partial gene of cytochrome C oxidase subunit I (COI) of wild silkmoth Cricula trifenestrata, and to detect the diagnostic sites from these gene for evaluation as species marker.

Increasing Cytosine Base Editing Scope and Efficiency With Engineered Cas9-PmCDA1 Fusions and the Modified sgRNA in Rice

open access: yesFrontiers in Genetics, 2019
Base editors that do not require double-stranded DNA cleavage or homology-directed repair enable higher efficiency and cleaner substitution of targeted single nucleotides in genomic DNA than conventional approaches.

First-principles study of high conductance DNA sequencing with carbon nanotube electrodes

open access: yes, 2011
Rapid and cost-effective DNA sequencing at the single nucleotide level might be achieved by measuring a transverse electronic current as single-stranded DNA is pulled through a nano-sized pore.

The effect of folate analogues and vitamin B12 on provision of thymine nucleotides for DNA synthesis in megaloblastic anemia [PDF]

open access: yesBlood, 1982
Abstract The role of vitamin B12 in the folate dependent biosynthesis of thymidine nucleotides is controversial. In an attempt to clarify this, three methods have been used to assess the relative efficacy of vitamin B12 (hydroxocobalamin) and various folate analogues in titrated concentrations at correcting ‘de novo’ thymidylate ...

Enhancing the Sensitivity of Colorimetric Aptasensor Through Cd2+‐Thymine Interactions in ssDNA Loops for Food Contaminant Detection

open access: yesFood Safety and Health, EarlyView.
This picture illustrates a novel aptamer‐based colorimetric assay, which demonstrates that high sensitivity, selectivity, and reliability for the rapid detection of cadmium ions (Cd2+) in water. Molecular dynamics (MD) simulations have revealed that the complexes are more stable, with Cd2+ binding more tightly to the thymine (T) within the aptamer loop.

Influence of a fluorobenzene nucleobase analogue on the conformational flexibility of RNA studied by molecular dynamics simulations [PDF]

open access: yes, 2006
Chemically modified bases are frequently used to stabilize nucleic acids, to study the driving forces for nucleic acid structure formation and to tune DNA and RNA hybridization conditions.
